参芪扶正汤对盆腔炎性疾病后遗症小鼠上生殖道炎症及粘连相关因子的影响

Effects of Shenqi Fuzheng decoction on upper genital tract inflammation and adhesion-related factors in mice with sequelae of pelvic inflammatory disease

目的研究参芪扶正汤对盆腔炎性疾病后遗症小鼠上生殖道炎性因子[肿瘤坏死因子-α(TNF-α)]及粘连相关因子[转化生长因子-β1(TGF-β1)、I型胶原蛋白(collagen I)]表达的影响。方法将60只雌性BALB/c小鼠随机分为6组,每组10只。除空白组外,其余组均采用宫颈注射盐酸/脂多糖法复制盆腔炎性疾病模型。建模成功后,阿司匹林组予阿司匹林肠溶片溶液灌胃,参芪扶正汤灌胃组、参芪扶正汤灌肠组分别予参芪扶正汤灌胃、灌肠,参芪扶正汤灌胃灌肠组给予参芪扶正汤灌胃及参芪扶正汤灌肠,空白组及模型组予羧甲基纤维素钠灌胃,各组均连续干预3个动情周期(每个4~5 d)。末次干预后第2天对各组小鼠进行脸谱疼痛评分(MGS),Western blot法检测各组小鼠上生殖道组织中TNF-α、TGF-β1、collagen I表达情况。结果模型组小鼠MGS评分及上生殖道组织中TNF-α、TGF-β1、collagen I相对表达量均明显高于空白组(P均<0.05)。各药物组小鼠MGS评分均明显低于模型组(P均<0.05),参芪扶正汤各组小鼠上生殖道组织中TNF-α、TGF-β1、collagen I相对表达量均明显低于模型组(P均<0.05),参芪扶正汤各组间MGS评分及TNF-α、TGF-β1、collagen I相对表达量比较差异均无统计学意义(P均>0.05)。结论参芪扶正汤可缓解盆腔炎性疾病后遗症所致的慢性盆腔痛,下调上生殖道组织中炎症因子及粘连相关因子的表达,且参芪扶正汤单一给药方式即可起到一定的治疗效果。

Objective It is to study the effects of Shenqi Fuzheng decoction on the expression of inflammatory factors[tumor necrosis factor-α(TNF-α)]and adhesion-related factors[transforming growth factor-β1(TGF-β1),collagen protein type I(collagen I)]in the upper genital tract of mice with sequelae of pelvic inflammatory disease.Methods Sixty female BALB/c mice were randomly divided into 6 groups with 10 mice in each group.Except for the blank group,the other groups were all replicated the pelvic inflammatory disease models by cervical injection of hydrochloric acid/lipopolysaccharide method.After successful modeling,the aspirin group was given aspirin enteric-coated tablet solution by gavage,the Shenqi Fuzheng decoction gavage group and the Shenqi Fuzheng decoction enema group were respectively given Shenqi Fuzheng decoction by gavage and enema,the Shenqi Fuzheng Decoction gavage and enema group was given Shenqi Fuzheng decoction by both gavage and enema,the blank group and the model group were administered with sodium carboxymethyl cellulose by intragastric administration.Each group was continuously intervened for 3 estrous cycles(4-5 days per cycle).On the 2nd day after the last intervention,the facial pain scores(MGS)of the mice in each group were evaluated,and the expressions of TNF-α,TGF-β1 and collagen I in the upper genital tract tissue of the mice in each group were detected by Western blot.Results The MGS scores and the relative expressions of TNF-α,TGF-β1 and collagen I in the upper genital tract tissue of the mice in the model group were significantly higher than those of the mice in the blank group(all P<0.05).The MGS scores of the mice in each treatment group were significantly lower than those of the mice in the model group(all P<0.05),and the relative expressions of TNF-α,TGF-β1 and collagen I in the upper genital tract tissue of the mice in each Shenqi Fuzheng decoction group were significantly lower than those of the mice in the model group(all P<0.05),there was no significant difference in the MGS score and the relative expression of TNF-α,TGF-β1 and collagen I among every Shenqi Fuzheng decoction group(all P>0.05).Conclusion Shenqi Fuzheng decoction can relieve chronic pelvic pain caused by sequelae of pelvic inflammatory disease,down-regulate the expression of inflammatory factors and adhesion-related factors in the upper reproductive tract tissue,and a single administration of this decoction can play a certain therapeutic effect.
