摘要
生物钟的形成与一系列生物钟基因及其调控基因构成的分子环路有关,其编码的蛋白可影响炎症细胞因子和趋化因子的表达,调控免疫细胞的增殖、分化和迁移,形成免疫功能的节律性变化。通过调控外界环境信号或基因修饰改变生物钟基因的表达,可诱发和加剧炎性肠病、类风湿性关节炎和1型糖尿病等多种自身免疫疾病。探讨生物钟基因的调控方式、固有免疫和适应免疫系统中生物钟的作用,以及生物钟基因在自身免疫性疾病发病机制和治疗中的潜在作用具有重要意义。
The formation of circadian clock is associated with the molecular loop composed of a series of core clock genes and their regulatory genes.The protein encoded by circadian clock genes can affect the expression of inflammatory cytokines and chemokines,regulate the proliferation,differentiation and migration of immune cells,and form the rhythmic fluctuations of immune function.Disrupting the expression of circadian clock genes by environmental signals or gene modifications,can induce and aggravate autoimmune diseases such as inflammatory bowel disease,rheumatoid arthritis and type 1 diabetes.It is of great significance to explore the modulation of circadian clock genes,the existence of circadian clock in innate immunity and adaptive immune system,and potential roles of circadian clock genes in the pathogenesis and treatment of autoimmune diseases.
