摘要
目的:探讨分泌型卷曲相关蛋白1(SFRP1)在人牙髓干细胞(hDPSCs)增殖、迁移及分化中的作用。方法:构建含敲低和过表达SFRP1的慢病毒,并将其转染至hDPSCs中,分别利用CCK-8法及细胞划痕实验检测hDPSCs增殖和迁移情况,茜素红染色检测矿化结节的形成,Western blot检测成牙/成骨蛋白和Wnt信号通路相关蛋白表达。结果:敲低SFRP1后hDPSCs的增殖、迁移能力增强,且DSPP、DMP1、ALP、OCN、Wnt3a、β-catenin和TCF1/LEF1表达水平上调,矿化结节数增加;过表达SFRP1后结果与之相反。结论:SFRP1抑制hDPSCs增殖、迁移及分化,其作用机制可能与Wnt/β-catenin信号通路相关。
Objective:To investigate the role of secreted frizzled related protein 1(SFRP1)in the proliferation,migration and differentiation of human dental pulp stem cells(hDPSCs).Methods:The lentivirus with knockdown and overexpression of SFRP1 gene were respectively constructed and transfected into hDPSCs.Cell proliferation and migration were respectively detected by CCK-8 and the wound-scratch assay.Alizarin red staining was used to detect mineralized nodules.Western blot was used to assess the protein expression of odontogenic/osteogenic proteins and Wnt signal pathway related proteins.Results:After knockdown of SFRP1,the ability of cell proliferation and migration were enhanced,and the expression of DSPP,DMP1,ALP,OCN,Wnt3a,β-catenin and TCF1/LEF1 were up-regulated.The formation of mineralized nodules increased.The results were opposite after overexpression of SFRP1 in the cells.Conclusion:SFRP1 inhibits the proliferation,migration and differentiation of hDPSCs,which may be related to the regulation of the Wnt/β-catenin signaling pathway.
