融合知识图谱探索川乌不同配伍的单酯型生物碱煎出率特性 被引量:3

Exploration on characteristics of decocting ratio of monoester aconitum alkaloid of Aconiti Radix in different compatibility by knowledge map

摘要 目的:基于知识图谱可视化挖掘川乌方药配伍特点,探讨川乌方药配伍的有效成分变化规律,为“药对-药性”关联研究提供基于数据挖掘指导下的物质基础变异特征的研究思路。方法:构建川乌方剂中的临床应用病证、配伍药物及药性的知识图谱,从药性角度出发,通过数据挖掘及临床用药经验选用白芍、桂枝、甘草作为川乌配伍研究对象;通过HPLC测定3种不同药物配伍川乌后汤液中的单酯型生物碱(苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱)含量变化,计算煎出率。结果:知识图谱显示,含川乌方剂在临床应用中主要治疗痹证和跌打损伤,治疗痹证时配伍的药物多为温性药,其次为平性、寒性和热性药;含量测定结果显示,与单煎液比较,川乌配伍白芍后3种单酯型生物碱煎出率显著升高(P<0.01,P<0.05),而配伍甘草后3种单酯型生物碱煎出率显著降低(P<0.01),配伍桂枝后3种单酯型生物碱煎出率差异无统计学意义。结论:川乌配伍不同药性药物后3种单酯型生物碱含量产生了差异性变化,体现了白芍增效、甘草减毒、桂枝协同的配伍特性,为川乌配伍临床应用提供了科学依据。 Objective:To explore the material basis of the medicinal effects of Aconiti Radix prescriptions,by the visualization of knowledge map and data mining of the prescriptions.Ultimately,the idea of‘drug pair-drug property’correlation research is provided based on the material basis variation characteristics of data mining.Method:To construct a knowledge map of the clinical applications,the compatibility of medicines and the medicinal properties in Aconiti Radix prescriptions,from the perspective of medicinal properties,Paeoniae Radix Alba,Cinnamomi Ramulus and Glycyrrhizae Radix et Rhizoma were selected as the subjects of Aconiti Radix pairings study by data mining and clinical experience of drug use.The content changes of monoester-type alkaloids(benzoyl neoaconitrile,benzoyl aconitine,and benzoyl hypoaconitrile)in decoctions of three different drugs by compatibility of Aconiti Radix were determined by HPLC,then the decocting ratio was calculated.Results:Knowledge mapping showed that Aconiti Radix prescriptions were mainly used in clinc to treat arthralgia syndrome and traumatic injury.The drugs used in the treatment of arthralgia syndrome were mostly warming drugs,followed by flat,cold and hot drugs.The content determination results showed that compared with the single decoction,the decoction rate of the three monoester-type alkaloids was significantly increased when Aconiti Radix was combined with Paeoniae Radix Alba(P<0.01,P<0.05),while the three monoestertype alkaloids were combined with licorice.The decoction rate of alkali was significantly decreased(P<0.01),and there was no statistically significant difference in the decoction rate of monoester alkaloids after combining with Cinnamomum cinnamon.Conclusion:The three monoester type alkaloid content of Aconiti Radix in combination with different medicinal drugs produced differential changes,reflecting the synergistic properties of Paeoniae Radix Alba to increase the effectiveness,Glycyrrhizae Radix et Rhizoma to reduce the toxicity and Cinnamomi Ramulus to synergize.This study provides scientific basis for the clinical application of Aconiti Radix through decocting ratio.
