国医大师张震疏调气机汤HPLC指纹图谱建立及7种成分含量测定

Establishment of HPLC fingerprint of Shutiao Qiji Decoction of TCM master ZHANG Zhen and determination of seven components

摘要 目的:建立不同产地的国医大师张震疏调气机汤HPLC指纹图谱,结合化学模式识别技术对疏调气机汤进行质量评价,同时对其7种成分进行含量测定。方法:Agilent Zorbax C_(18)柱(250 mm×4.6 mm,5μm);乙腈-0.1%磷酸水溶液,梯度洗脱;检测波长:280 nm;流速:1 mL/min;进样量:10μL;柱温:25℃。结果:15批疏调气机汤的HPLC指纹图谱有19个共有峰,相似度在0.965~1.000,通过与混合对照品比对,指认出芍药苷、甘草苷、阿魏酸、柚皮苷、橙皮苷、新橙皮苷、柴胡皂苷B_(2)7种化学成分。定量分析条件方法学考察结果良好,7个成分加样回收率在93.72%~102.11%;15批样品中芍药苷、甘草苷、阿魏酸、柚皮苷、橙皮苷、新橙皮苷、柴胡皂苷B2的质量分数分别为3.0572~8.2173、0.0994~0.2308、0.2343~0.4172、1.5538~2.6403、0.2244~0.4323、0.7516~1.7868、4.4271~9.6341 mg/g。结论:本研究建立的HPLC指纹图谱结合化学模式识别的方法稳定、可靠、简便准确,可以更系统、全面地为疏调气机汤的质量控制评价提供依据。 Objective:To establish the HPLC fingerprint of Shutiao Qiji Decoction of TCM master ZHANG Zhen from different production areas,combine with chemical pattern recognition technology to evaluate the quality of Shutiao Qiji Decoction,and determine the content of 7 components.Methods:Agilent Zorbax C_(18) column(250 mm×4.6 mm,5μm);acetonitrile-0.1%phosphoric acid aqueous solution,gradient elution;Detection wavelength:280 nm;flow rate:1 mL/min;injection volume:10μL;column temperature:25℃.Results:The HPLC fingerprint of Shutiao Qiji Decoction had 19 common peaks,and the similarity was between 0.965 and 1.000.By comparing with the mixed reference substance,seven chemical constituents including paeoniflorin,liquiritin,ferulic acid,naringin,hesperidin,neohesperidin and saikosaponin B_(2) were identified.The quantitative analysis conditions and methodological investigation results were good,and the recovery rate of 7 components was 93.72%~102.11%.The mass fractions of paeoniflorin,liquiritin,ferulic acid,naringin,hesperidin,neohesperidin and saikosaponin B_(2) in the 15 batches of samples were 3.0572~8.2173,0.0994~0.2308,0.2343~0.4172,1.5538~2.6403,0.2244~0.4323,0.7516~1.7868,4.4271~9.6341 mg/g,respectively.Conclusion:The method of HPLC fingerprint and chemical pattern recognition established in this study is stable,reliable,simple and accurate,and can provide a basis for the quality control evaluation of Shutiao Qiji Decoction more systematically and comprehensively.
