艾灸胃经穴对慢性萎缩性胃炎大鼠胃黏膜miR-146a、NF-κBp65及COX-2表达的影响 被引量:4

Effects of moxibustion on points of stomach meridian on the expression of miR-146a,NF-κBp65 and COX-2 in gastric mucosa of rats with chronic atrophic gastritis

摘要 目的:探讨艾灸胃经穴对慢性萎缩性胃炎(CAG)大鼠胃黏膜组织miR-146a、NF-κBp65及COX-2表达的影响。方法:50只S D大鼠随机分为正常组、模型组、艾灸正常组、艾灸组、艾灸非穴组,每组10只。采用氨水溶液和脱氧胆酸钠交替饮水结合饥饱失常法制备C A G模型。干预组在造模完成后进行艾灸干预,HE染色观察胃黏膜组织形态;Western Blot检测NF-κBp65和COX-2蛋白表达水平;Q-PCR检测miR-146a、NF-κBp65、COX-2 mRNA表达水平。结果:与正常组比较,模型组大鼠胃黏膜中腺体萎缩明显,艾灸正常组无明显变化,艾灸胃经穴则改善了胃黏膜组织病理结构;模型组中miR-146a mRNA含量显著降低(P<0.05);模型组NF-κBp65、COX-2 mRNA及蛋白水平均显著升高(P<0.01,P<0.05)。与模型组比较,艾灸组中miR-146a mRNA表达显著上调(P<0.01),NF-κBp65 mRNA含量和蛋白表达水平显著降低(P<0.01)。结论:艾灸胃经穴可能通过调节相关炎症因子起到治疗CAG的作用。 Objective:To explore the effects of moxibustion on the points of stomach meridian on the expression of chronic atrophic gastritis(CAG)miR-146a,NF-κBp65 and COX-2 in the gastric mucosa of rats.Methods:Fifty SD rats were randomly divided into normal group,model group,normal moxibustion group,moxibustion group,and moxibustion non-acupoint group,with 10 rats in each group.CAG model was prepared by using ammonia solution and sodium deoxycholate alternately and freely drinking combined with hunger and satiety.Each group began to moxibustion after the completion of the model.The morphology of gastric mucosa was observed by HE staining;The level of nuclear factor-κBp65(NF-κBp65)and cyclooxygenase 2(COX-2)was detected by Western Blot.The miR-146a,NF-κBp65,COX-2 mRNA expression level were detected by Q-PCR.Results:Compared with the normal group,the glands in the gastric mucosa of the model group were significantly atrophy,while the moxibustion normal group had no obvious changes.Moxibustion on the stomach meridian points improved the pathological structure of the gastric mucosa,the miR-146a mRNA expression in the model group was significantly decreased(P<0.01),the NF-κBp65,COX-2 mRNA content and the protein levels of the model group were significantly higher(P<0.01,P<0.05).Compared with the model group,the miR-146a mRNA expression in the moxibustion group was significantly up-regulated(P<0.01),and the NF-κBp65 mRNA content and protein expression level were significantly reduced(P<0.01).Conclusion:Moxibustion on the points of the stomach meridian can treat CAG by improving mucosal inflammation in rats.
