摘要
目的:观察意乐藏香(柱香和陀香)燃烧不同时间对模式菌株和空气微生物的生长抑制作用。方法:利用菌落计数法计算香薰对模式菌株大肠杆菌和金黄色葡萄球菌生长抑制情况,同时对空气中微生物进行培养、纯化、DNA提取、16S rRNA基因的扩增与测序分析,探讨香薰对空气微生物抑制效果和微生物种类变化。结果:两种藏香对模式菌株大肠杆菌无显著抑制效果,对金黄色葡萄球菌燃香1.5 h后,最佳抑制效果达到70%以上。空气微生物抑制实验表明,两种藏香对空气细菌的抑制效果明显,燃香1.5 h后,空气中可培养细菌抑菌率达80%以上。燃香前实验室可培养微生物共包含4个门:放线菌门(Actinobacteria)、拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)、变形菌门(Proteobacteria);5个纲,11个目,13个科,19个属,38个种。随着藏香燃烧时间的延长,微生物数量和种类数减少,点燃1.5 h后,只剩下厚壁菌门的7个种。结论:意乐藏香(陀香)对模式菌株金黄色葡萄球菌有明显抑制作用;2种意乐藏香对空气中微生物的生长具有明显抑制作用,可以起到良好的杀菌效果。
Objective:To analyze the inhibition rate of two Tibetan incenses on pattern and aerial microorganism under different length of burning time.Methods:The inhibition rate of two Tibetan incenses burning for Escherichia coli and Staphylococcus aureus were calculated,at the same time,the air microorganism inhibition effect and changes of microbial species based on purification,DNA extraction,16S rRNA gene amplification and sequencing were researched.Results:Burning two kinds of Tibetan incenses had significant inhibition effect on S.aureus and inhibition rate above 70%after burning one and half hours,burning two kinds of Tibetan incenses had no significant inhibition effect on E.coli.The aerial microorganism inhibition results show that the effect was obvious of burning two Tibetan incenses and inhibition rate above 80%after burning one and half hours.Cultivable microorganisms were distributed in 4 phyla(Actinobacteria,Bacteroidetes,Firmicutes,Proteobacteria),5 classes,11 orders,13 families,19 genera,38 species before burning Tibetan incenses.As the burning time prolonged,microbial populations and species decreased,after burning one and half hours only had 7 species in Firmicutes.Conclusion:Yile Tibetan incense(tuo shap)has obvious inhibitory effect on model strain Staphylococcus aureus,and the two kinds of Tibetan incense have obvious inhibitory effect on the growth of microorganisms in the air,which have a good bactericidal effect.
