一种高效的藻类叶绿素定量提取方法

摘要 叶绿素的定量检测对于藻类生物量的估算非常重要。当前常用丙酮和乙醇作为叶绿素提取溶剂,但对于某些藻类提取效果不佳。研究表明,二甲基甲酰胺(DMF)作为提取溶剂比丙酮和乙醇更有效。本研究以蛋白核小球藻为实验材料,以加热作为辅助措施,对比研究了DMF、丙酮以及乙醇这三种有机溶剂的叶绿素提取效率。结果表明,热DMF比热丙酮和热乙醇具有更高的提取效率;在90℃温度下,用DMF浸泡藻细胞沉淀5 min可获得最佳提取率。改进的热DMF法提取效率高,操作耗时短,精密度高,特别适合处理具有叶绿素提取抗性的藻类材料。 The quantitative detection of chlorophyll is very important for the estimation of algae biomass.At present,acetone and ethanol are commonly used as chlorophyll extraction solvents,but for some algae,the extraction effect is not good.Studies have shown that dimethylformamide(DMF)is more effective as an extraction solvent than acetone and ethanol.In this study,using Chlorella phyrenoidosa as the experimental material and heating as an auxiliary measure,the chlorophyll extraction efficiency of three organic solvents,DMF,acetone and ethanol,were compared.The results show that hot DMF has a higher extraction efficiency than hot acetone and hot ethanol,the best extraction rate can be obtained by soaking the algal cell pellet in DMF for 5 min at a temperature of 90℃.The improved hot DMF method has high extraction efficiency,short operation time and high precision,and is particularly suitable for processing algae materials with chlorophyll extraction resistance.
