摘要
目的探究大黄素对去势绝经小鼠甘油通道水通道蛋白7(AQP7)表达的组织特异性的影响。方法选取50只雌性C57BL/6小鼠,平均分为空白对照组、模型组和大黄素低、中、高组,除空白对照组,其余各组均构建去势绝经小鼠模型。测定5组小鼠血清性激素水平[雌二醇(E2)、孕酮(P)、促卵泡生成素(FSH)、促黄体生成素(LH)]、卵巢指数、子宫指数和子宫内膜厚度、不同组织中[性腺脂肪(g-Fat)、腹股沟脂肪(i-Fat)、肾周脂肪(p-Fat)]AQP7 mRNA和蛋白的表达水平、雌激素受体基因ERα和ERβmRNA水平,同时分析血清性激素对小鼠模型AQP7表达的影响。结果与空白对照组比较,其余4组E2、P、卵巢指数、子宫指数、子宫内膜厚度、雌激素受体基因ERα和ERβmRNA水平均降低,FSH、LH、不同组织中AQP7 mRNA和AQP7蛋白水平均升高,且模型组最显著,差异均有统计学意义(P<0.05)。除空白对照组外,其余3个药物组与模型组比较,E2、P、卵巢指数、子宫指数、子宫内膜厚度、雌激素受体ERα和ERβmRNA水平均降低,FSH、LH、不同组织中AQP7 mRNA和蛋白均升高,差异有统计学意义(P<0.05)。大黄素高、中、低剂量组上述指标比较,差异均有统计学意义(P<0.05)。E2、P、FSH、LH与g-Fat和p-Fat中AQP7呈正相关(P<0.05)。FSH与i-Fat中的AQP7呈负相关(P<0.05)。结论大黄素能够通过改善去势绝经小鼠的血清性激素水平从而降低不同组织中甘油通道水通道蛋白7的表达,对小鼠卵巢和子宫具有一定的保护作用。
Objective To investigate the effects of emodin on the tissue specificity of glycerol channel aquaporin 7(AQP7)expression in castrated menopausal mice.Methods Fifty female C57BL/6 mice were randomly divided into blank control group,model group and low-dose,medium-dose and high-dose emodin groups.Except for the mice in blank control group,the animal models with castrated menopause were established in the mice in the other groups.The serum levels of sex hormones(E2,P,FSH,LH),ovarian index,uterine index and endometrial thickness,and expression levels of AQP7 mRNA and protein in different tissues,and estrogen receptor gene ERαand ERβmRNA levels were detected,moreover,the effects of serum sex hormones on the expression of AQP7 was analyzed.Results Compared with those in blank control group,the levels of E2,P,ovary index,uterine index and endometrial thickness,and expression levels of ERαand ERβmRNA in the other four groups were significantly decreased,however,the levels of FSH,LH,and expression levels of AQP7 mRNA and protein in different tissues were significantly increased,moreover,the changes were most significant in model group(P<0.05).And the levels of E2,P,ovary index,uterine index and endometrial thickness,and expression levels of ERαand ERβmRNA in the three emodin groups were significantly lower than those in model group,however,the levels of FSH,LH,and expression levels of AQP7 mRNA and protein in different tissues were significantly high than those in model group(P<0.05).Moreover there were significant differences in above indexes among the three emodin groups(P<0.05).In addition there was a positive correlation between serum sex hormones and the expression of AQP7 in g-Fat and p-Fat(P<0.05),and there was a negative correlation between the expression of AQP7 in i-FAT and FSH(P<0.05).Conclusion Emodin can reduce the expression of AQP7 in different tissues by improving the serum levels of sex hormones in castrated menopausal mice,which has a certain protective effect on the ovary and uterus of mice.
