三相萃取法分离纯化黄晶果多糖的工艺优化及抗氧化性 被引量:2

Optimization of Extraction Process by Three-Phase Extraction and Antioxidant Activity of Polysaccharides from Abiu.

摘要 为快速实现多糖分离同时避免其醇沉过程,采用三相萃取法分离纯化黄晶果多糖,研究多糖的萃取工艺及体外抗氧化活性。以黄晶果为原料,通过单因素、正交试验方法优化多糖萃取工艺参数;通过Fenton反应法、DPPH法和ABTS法评价其抗氧化能力。结果表明:最佳工艺条件为硫酸铵质量分数为30%、叔丁醇添加量为15 mL、萃取温度为45℃,此条件下黄晶果多糖萃取率为79.55%,蛋白质去除率为55.03%,黄酮提取率为8.38%;黄晶果多糖对·OH、DPPH·和ABTS自由基的半数清除率IC_(50)值分别为1.85、1.88、2.69 mg/mL。研究结果表明黄晶果多糖的体外抗氧化能力较强,该工艺较好地保留了黄晶果多糖的活性。 The three phase extraction method can avoid the alcohol precipitation process of polysaccharides to separate polysaccharides in a short time.The extraction and antioxidant activity of polysaccharides from Abiu.were investigated in this work.Abiu.was used as the experimental material,and the extraction process of polysaccharides from Abiu.was optimized by single factor and orthogonal test.The antioxidant activities of Abiu.polysaccharides were tested by Fenton’s oxidation reaction,DPPH assay and ABTS assay.The optimum extraction conditions were as follows:(NH)SOaddition 30%,tertbutyl alcohol addition 15 mL,extraction temperature 45℃,and the extraction rate of polysaccharides reached 79.55%,the protein extraction rate reached 55.03%,and the extraction rate of flavonoids reached 8.38%.The antioxidant activity of Abiu.polysaccharides was studied.The IC_(50) values of the Abiu.polysaccharides to OH,DPPH and ABTS radicals were 1.85 mg/mL,1.88 mg/mL and 2.69 mg/mL respectively.The results showed that Abiu.polysaccharides had strong antioxidant activity,and the three-phase extraction process also retained its activity well.
