摘要
目的 观察自噬相关基因微管相关蛋白1轻链3(microtubule-associated protein 1 light chain 3,LC3)在神经母细胞瘤(neuroblastoma, NB)中的表达,探讨其在NB发生、发展过程中所起的作用。方法 收集经哈尔滨医科大学附属哈尔滨市儿童医院病理证实为NB的手术标本68例,通过免疫组化法检验LC3在NB组织与瘤旁组织内的表达,同时以NB患者的临床资料为基础,研究LC3的表达与NB临床病理学特征以及患者预后的关系。结果 免疫组织化学染色结果显示LC3蛋白在NB细胞浆中呈阳性表达,而瘤旁组织中LC3表达为阴性;LC3蛋白在NB高分期(Ⅲ~Ⅳ期)肿瘤组织内的表达水平明显高于NB低分期(Ⅰ~Ⅱ期)肿瘤组织(P<0.05);同时在有淋巴结转移组的表达水平明显高于无淋巴结转移组(P<0.05);而LC3蛋白在NB组织中的表达水平与性别、年龄、大小、肿瘤部位、病理分型等参数无显著相关性(P>0.05);LC3蛋白的高表达将导致NB患者预后较差。结论 LC3蛋白于NB组织内表达水平显著,其表达水平和NB的肿瘤临床分期、是否有淋巴结转移存在相关性,同时和NB患者的预后密切相关。
Objective To observe the expression of the microtubule-associated protein 1 light chain 3(LC3) in neuroblastoma(NB), and discusse its role in occurrence and development of neuroblastoma. Methods Sixty-eight surgical specimens through pathologically confirmed NB from Harbin Children’s Hospital Affiliated to Harbin Medical University were collected. The expression of LC3 in NB and paralumoral cell tissues was detected by immunohistochemistry, and the relationship of LC3 expression with clinicopathological characteristics and prognosis of NB was studied based on the clinical data. Results Immunohistochemical staining results showed that LC3 was positively expressed in the cytoplasm, and LC3 was negatively expressed in the para-tumor tissues. Statistical analysis showed that the expression levels of LC3 in high stage tumors(Ⅲ~Ⅳ stage) were higher than those in low stage tumors(Ⅰ~Ⅱ stage)(P<0.05). The expression level in the group with lymph node metastasis was significantly higher than that in the group without lymph node metastasis(P<0.05). High expression of LC3 lead to poor prognosis in NB patients. Conclusion The expression level of LC3 protein is significant in NB tissues, which is correlated with tumor clinical stage of NB and whether there is lymph node metastasis, and is closely related to the prognosis of NB patients.
