摘要 N,N-二甲基甲酰胺(DMF)是一种优良的工业溶剂和有机合成材料,它广泛应用于制革、化工、医药、农药等各个生产行业。含DMF的废水毒性大,对环境造成严重的危害。筛选分离得到能够高效降解DMF的菌株,为含DMF污水的处理提供专用菌剂,保证污水处理后达标排放。采用富集和选择培养的方法,以山东某环保公司的活性污泥为材料进行DMF降解菌的筛选,对降解菌的形态特征、生理生化特性和DMF降解率进行测定,并且采用16S rDNA对降解菌进行鉴定,分析其对DMF的降解特性,并对该菌株在相关水质方面的应用进行了研究。分离筛选到一株能以DMF为唯一碳源的降解菌,经16S rDNA鉴定,该菌株D1为惠氏副球菌(Paracoccuswyeth),其具有较强的DMF降解能力及耐受性,能够实现一定程度对DMF污染物的去除,这为今后此菌剂在不同水质中的应用奠定了基础。 N,N-dimethylformamide(DMF) is an excellent industrial solvent and organic synthetic material. It is widely used in leather, chemical, pharmaceutical, pesticide and other industries. The wastewater containing DMF is highly toxic and causes serious harm to the environment. The strain which could efficiently degrade DMF was screened and isolated, in order to provide a special bacterial agent for treatment of wastewater containing DMF and ensure that the sewage is discharged up to the standard after treatment. The DMF degrading bacteria were screened from activated sludge of an environmental-protection company in Shandong province by enrichment culture and selective culture. The DMF degrading bacteria were identified by morphological characteristics, physiological,biochemical characteristics and 16S rDNA. Moreover, the DMF degradation rate of the bacteria was determined. The application of this strain was studied. The results showed that, a strain of DMF degradation bacteria was isolated and screened, which could utilize DMF as the sole carbon. The strain D1 was identified as Paracoccuswyeth by 16S rDNA.The strain showed strong ability of DMF degradation and tolerance, and could realize the removal of DMF pollutants to a certain extent, which could lay a foundation for the future application of this bacterial agent in different water quality.
