HPLC法测定三拗片中4个成分的含量

摘要 目的建立测定三拗片中夏佛塔苷、甘草苷、异甘草苷、甘草酸含量的HPLC法。方法采用Ultimate XB-C_(18)色谱柱(250 mm×4.6 mm,5μm);VWD紫外多波长检测器;流动相为体积分数为0.2%的磷酸水(A)-乙腈(B)溶液,梯度洗脱;流速为1 mL·min^(-1);检测波长为254 nm;进样量为10μL^(-1)柱温为30℃。结果夏佛塔苷、甘草苷、异甘草苷、甘草酸分别在质量浓度0.5~20.2μg·L^(-1)、242.4~2816.6 mg·L^(-1)、1.8~32.2μg·L^(-1)和171.1~5359.2 mg·L^(-1)内与峰面积呈良好线性关系,相关系数r分别为0.9990、0.9995、0.9993、0.9990,平均回收率分别为93.82%、106.6%、109.8%和99.94%(n=9)。RSD分别为1.6%、1.9%、1.6%和2.7%,精密度、重复性、稳定性及耐用性的RSD均小于2.0%。结论本方法可用于三拗片中夏佛塔苷、甘草苷、异甘草苷和甘草酸的含量测定。 Objective To establish a HPLC method for the determination of schaftoside,liquiritin,isoliquiritin and glycyrrhetinic acid in SanAaoPian.Methods An Ultimate XB-C_(18)column(250 mm×4.6 mm,5μm)with VWD detector was used.The mobile phase consisted of 0.2%phosphoric acid aqueous solution(A)-acetonitrile(B)with gradient elution.The flow rate was 1 mL·min^(-1);the detection wavelength was 254 nm;the injection volume was 10μL^(-1)the column temperature was 30℃.Results The peak areas of schaftoside,liquiritin,isoliquiritin and glycyrrhetinic acid showed good linear relationships with their concentrations in the ranges of 0.5-20.2μg·L^(-1),242.4-2816.6 mg·L^(-1),1.8-32.2μg·L^(-1)and 171.1-5359.2 mg·L^(-1),respectively.The correlation coefficients r were 0.9990,0.9995,0.9993 and 0.9990,the mean recoveries were 93.82%,106.6%,109.8%and 99.94%(n=9),and the RSDs were 1.6%,1.9%,1.6%and 2.7%,respectively.The RSDs for precision,reproducibility,stability and durability were less than 2.0%.Conclusion The method can be used for the determination of schaftoside,liquiritin,isoliquiritin and glycyrrhetinic acid in SanAoPian.
