京尼平苷调控Wnt/β-cantenin信号通路抑制胃癌转移的机制研究 被引量:2

摘要 目的探讨京尼平苷(GEN)抑制人胃癌SGC-7901细胞转移的作用及其可能机制。方法培养人胃癌SGC-7901细胞,使用GEN处理细胞,通过CCK8法检测GEN对SGC-7901细胞增殖的影响,运用Transwell实验检测GEN对SGC-7901细胞迁移和侵袭能力的作用,运用Westernblot和q-PCR检测GEN对SGC-7901细胞β-catenin、E-cadherin、Vimentin和Snail蛋白及mRNA表达的作用;运用Wnt通路激活剂(LiCl)和GEN联合处理SGC-7901细胞后,观察对SGC-7901细胞迁移和侵袭能力的影响,对β-catenin、E-cadherin、Vimentin和Snail蛋白及mRNA表达的影响。结果Transwell实验结果显示,GEN能够抑制SGC-7901细胞的迁移和侵袭能力下降,差异有统计学意义(P<0.05)。Westernblot和q-PCR结果表明,GEN能够促进E-cadherin的表达,抑制β-catenin、Vimentin和Snail的表达,差异有统计学意义(P<0.05)。与GEN组比较,Wnt激活剂LiCl与GEN联合处理会促进胃癌SGC-7901细胞的迁移和侵袭,促进β-catenin、Snail和Vimentin蛋白及mRNA表达,抑制E-cadherin蛋白及mRNA表达,差异有统计学意义(P<0.05)。结论GEN可能通过抑制Wnt/β-catenin信号通路而抑制胃癌SGC-7901细胞迁移和侵袭。 Objective To investigate the effect of Geniposide(GEN)on the metastasis of human gastric cancer SGC-7901 cells and its possible mechanism.Methods Human gastric cancer SGC-7901 cells were cultured and treated with GEN.The influence of GEN on the proliferation of SGC-7901 cells were measured by Cell Counting Kit-8(CCK-8).The influence of GEN on the migration and invasion of SGC-7901cells were detected through Transwell assay.Westernblot and q-PCR were used to detect the effect of GEN on the expression ofβ-catenin,E-cadherin,Vimentin and Snail protein and mRNA in SGC-7901 cells.Wnt pathway activator(LiCl)and GEN were used to observe the effects on the migration and invasion of SGC-7901 cells,and on the expression ofβ-catenin,E-cadherin,Vimentin and Snail protein and mRNA.Results The results of the Transwell assay showed that GEN can inhibit the migration and invasion of SGC-7901 cells,with statistical significance(P<0.05).Westernblot and q-PCR results showed that GEN can promote the expression of E-cadherin and inhibit the expression ofβ-catenin,Vimentin and Snail,with statistical significance(P<0.05).Compared with the GEN group,the combined treatment of LiCl and GEN can promote the migration and invasion of gastric cancer SGC-7901 cells,with statistical significance(P<0.05),promoteβ-catenin,Snail and Vimentin protein and mRNA expression,and inhibit E-cadherin protein and mRNA expression(P<0.05).Conclusion GEN may inhibit the migration and invasion of gastric cancer SGC-7901 cells by inhibiting the Wnt/β-catenin signaling pathway.
