黄连素对人肺癌细胞增殖、凋亡、迁移、侵袭能力的影响

Effect of berberine on proliferation, apoptosis, migration, and invasion of lung cancer cells

目的 探讨黄连素对肺癌细胞的增殖、凋亡、迁移和侵袭的影响。方法 用0~100μmol/L黄连素处理人肺癌细胞株24 h后,用MTT法检测细胞增殖,流式细胞术检测细胞凋亡,划痕实验检测细胞迁移,Transwell实验检测细胞侵袭,蛋白质印迹检测相关蛋白表达。结果 黄连素显著抑制肺癌细胞增殖、迁移、侵袭,促进细胞凋亡(P<0.01);上调Bad、Caspase-3、Caspase-9、TIMP-1表达,下调Bcl-2、MMP-2表达。结论 黄连素可抑制增殖、迁移、侵袭能力,促进肺癌细胞凋亡。

Objective To investigate the effect of berberine on proliferation, apoptosis, migration, and invasion of lung cancer cells. Methods After treated with 0-100 μmol/L berberine for 24 hours, proliferation, apoptosis, migration, invasion,and associated protein expression of human lung cancer cell lines were detected by MTT, flow cytometry, scratch, Transwell,and Western blot, respectively. Results Berberine significantly inhibited cell proliferation, migration and invasion, but enhanced apoptosis of lung cancer cells(P<0.01);and upregulated Bad, Caspase-3, Caspase-9, and TIMP-1 expression, but downregulated Bcl-2 and MMP-2 expression. Conclusion Berberine inhibites proliferation, migration and invasion, and promotes apoptosis of lung cancer cells.
