摘要
目的探讨miR-506-3p对肝细胞的增殖、迁移与侵袭的作用及机制。方法RT-qPCR法检测肝癌组织和肝癌细胞中的miR-506-3p表达,MTT法和EdU掺入法研究过表达miR-506-3p对肝癌细胞增殖的影响,划痕法和Transwell小室法评估过表达miR-506-3p对肝癌细胞迁移和侵袭的影响,荧光素酶报告基因测定法和功能修复实验法确定miR-506-3p是否靶向COPB2 mRNA的3’-非翻译区(3’-UTR)发挥作用。结果miR-506-3p在肝癌组织和肝癌细胞中低表达;miR-506-3p过表达可抑制肝癌细胞增殖、迁移和侵袭;miR-506-3p通过直接结合COPB2 mRNA的3’-UTR负调控COPB2的表达,过表达COPB2能逆转miR-506-3p过表达对肝癌细胞的增殖、迁移和侵袭的抑制作用。结论miR-506-3p通过靶定COPB2 mRNA来抑制肝癌细胞的增殖、迁移和侵袭。
Objective To explore the effect and mechanism of miR-506-3p on the proliferation,migration and invasion of hepatoma carcinoma cells.Methods RT-qPCR was used to detect the expression of miR-506-3p in liver cancer tissues and liver cancer cells;MTT method and EdU incorporation method were used to study the effect of overexpression of miR-506-3p on the proliferation of liver cancer cells;scratch method and Transwell chamber method were used to evaluate the effect of overexpression of miR-506-3p on the migration and invasion of liver cancer cells;luciferase reporter gene assay and functional repair experiments were used to determine whether miR-506-3p targeted the 3’-untranslated region(3’-UTR)of COPB2 mRNA.Results miR-506-3p was lowly expressed in liver cancer tissues and liver cancer cells;overexpression of miR-506-3p inhibited the proliferation,migration and invasion of hepatoma cells;miR-506-3p negatively regulated the expression of COPB2 by directly binding to the 3’-UTR of COPB2 mRNA.Overexpression of COPB2 could reverse the inhibitory effect of miR-506-3p overexpression on proliferation,migration and invasion of hepatoma cells.Conclusion miR-506-3p inhibits the proliferation,migration and invasion of hepatoma cells by targeting COPB2 mRNA.
