摘要 乳腺癌患者死亡率居高不下的主要原因在于肿瘤的复发,而复发的关键因素是多重耐药性(multidrug resistance,MDR)的产生并影响治疗效果和预后。MDR的产生涉及多种分子机制,包括外排转运蛋白表达的增加(如P-糖蛋白、乳腺癌耐药蛋白或多重耐药相关蛋白)、上皮间质转化、乳腺癌干细胞的抗性发展等。本文讨论了乳腺癌目前的分类及病理生理特性,重点阐述了乳腺癌耐药、转移和复发的潜在机制。 The main reason for the high mortality in breast cancer patients is the recurrence of the tumor,and the key factor for recurrence is the generation of multidrug resistance(MDR),which affects the efficacy and prognosis of treatment.Multiple molecular mechanisms may be involved in the generation of MDR,including increased expression of efflux transporters(e.g.,P-glycoprotein,breast cancer drug resistance protein,or multidrug resistance-associated protein),epithelial-mesenchymal transition,resistance to breast cancer stem cells sexual development and more.This review discusses the current classification and pathophysiological properties of breast cancer,focusing on the mechanisms underlying breast cancer drug resistance,metastasis,and recurrence.
