摘要
传统病原学检测方法对于兽医临床感染病原的鉴定具有一定的局限性,尽管高通量测序技术极大地提高了未知病原感染的检测水平,但是对于无DNA复制周期的RNA病毒的捕获,仍存在一定难度。基于此,本研究利用RNA建库技术对1例猫传染性腹膜炎的腹水与血清样本进行了高通量测序,并对所得数据进行分析以判断感染病原,结果提示病猫腹水样本中携带猫传染性腹膜炎病毒(FIPV)、猫星状病毒(FeAstV)与犬细小病毒(CPV),血清样本中携带猫传染性腹膜炎病毒(FIPV),其中,FIPV与FeAstV为正链RNA病毒,CPV为单链DNA病毒。总之,本研究不仅证实基于RNA建库的高通量测序技术能够同时检测到DNA与RNA病毒混合感染,而且能够为动物未知病原感染的检测提供新思路与参考。
It is well known that traditional pathogen detection methods have certain limitations in the identification of pathogen in veterinary clinical infection.High-throughput sequencing has greatly improved the detection spectrum of pathogen,but it is still difficult to capture RNA viruses which produce only RNA fragments during replication.This study focused on RNA library construction technology to sequence the ascites and serum samples from a case of a cat with feline infectious peritonitis,and analyzed the data to identify the causative pathogen.The results showed that feline infectious peritonitis virus(FIPV),feline astrovirus(FeAstV)and canine parvovirus(CPV)were simultaneously detected in the sequencing data of serum and blood samples from the diseased cat.FIPV and FeAstV are RNA viruses belonging to orthornavirae,and CPV is a member of monodnaviria with a genome of single-stranded DNA.In short,this study demonstrated that high-throughput sequencing based on RNA library construction technology can simultaneously detect DNA and RNA virus infections in one sample,the research also provides new methods and references for the detection of unknown pathogen infection in animals.
