镉对鸭破骨细胞焦亡的影响 被引量:1

Effect of cadmium on pyroptosis of duck osteoclast

摘要 为探讨镉暴露对鸭破骨细胞焦亡的影响,本试验用不同浓度的镉(0,5和10μmol/L)处理鸭破骨细胞12 h。通过显微镜观察破骨细胞形态变化,ELISA试剂盒检测IL-1β和IL-18的含量,流式细胞术检测破骨细胞焦亡,Hoechst和PI染色观察细胞核数量变化。结果显示,镉暴露后鸭破骨细胞表现出明显的形态改变,主要表现为细胞肿胀,细胞膜可见大量孔洞。与对照组相比,镉暴露明显增加了鸭破骨细胞内LDH的释放,细胞因子IL-1β和IL-18的产生也显著升高(P<0.01)。Caspase-1的活性较对照组显著增加(P<0.01),FITC和PI双染的阳性破骨细胞数也明显升高(P<0.01)。Hoechst和PI染色结果表明,镉暴露明显增加了红色和蓝色荧光强度。上述结果表明,镉暴露可诱导鸭破骨细胞发生焦亡。 In order to investigate the effects of cadmium exposure on the pyroptosis of duck osteoclasts,different concentrations of cadmium(0,5,10μmol/L)were used to treat duck osteoclasts for 12 h.The morphological change of osteoclasts was observed by microscope,the contents of IL-1β and IL-18 were detected by ELISA kit,the pyroptosis of osteoclasts was detected by flow cytometry,and the changes of cells nuclear were observed by Hoechst and PI staining.The results showed that the osteoclasts of ducks showed obvious morphological changes after cadmium exposure,which mainly manifested cell swelling and a large number of holes in the cell membrane.Compared with the control group,cadmium exposure significantly increased the release of LDH and the production of cytokines IL-1β and IL-18 in duck osteoclasts(P<0.01),the activity of caspase-1 was significantly increased,and the number of positive osteoclasts double stained with FITC and PI was also significantly increased(P<0.01).Hoechst and PI staining results showed that the fluorescence intensity of red and blue significantly increased under cadmium exposure.These results indicate that cadmium exposure can induce the pyrotosis of duck osteoclasts.
