摘要
以麻黄雏鸡为研究对象,采用脂多糖(LPS)联合D-氨基半乳糖(GalN)诱导雏鸡肝损伤模型,评价杜鹃素对肝损伤的防治作用,为杜鹃素在兽医临床上的广泛应用提供理论基础和实验依据。将1日龄雏鸡随机分为5组(n=7):空白组、LPS/GalN模型组、LPS/GalN+杜鹃素(低、中、高剂量)组,适应环境3 d,在第4,6,8天灌胃给药杜鹃素,空白组和LPS/GalN组灌胃相同剂量生理盐水。第8,9,10天腹腔注射LPS(250μg/kg)和GalN(250 mg/kg)。24 h后收取血清和肝脏。每日称重,检测血清中ALT、AST的活性和肝脏中SOD、GSH-PX的水平,HE染色法检查肝脏组织病理学变化,qRT-PCR法检测肝脏中TNF-α、IL-1β、iNOS、COX2 mRNA的相对表达量。结果显示,与空白组相比,LPS/GalN组增重率显著降低,杜鹃素预处理组增重率高于模型组;给予LPS/GalN后,血清中ALT、AST活性极显著高于空白组,杜鹃素预处理组可以极显著降低血清中ALT、AST活性;肝组织病理学检查结果表明,与空白对照组相比,给予LPS/GalN后,肝细胞结构消失、肝细胞索紊乱并且有炎性细胞浸润,而中剂量和高剂量杜鹃素组发现,雏鸡肝脏中肝细胞结构趋于正常,炎性细胞减少;与空白组相比,LPS/GalN模型组肝脏中SOD、GSH-PX水平极显著降低,而TNF-α、IL-1β、iNOS、COX2 mRNA的表达量极显著升高;与LPS/GalN模型组相比,杜鹃素预处理组能显著提高SOD、GSH-PX的活性,并极显著降低TNF-α、IL-1β、iNOS、COX2 mRNA的表达量。综上,LPS/GalN成功建立了雏鸡肝损伤模型,杜鹃素可以通过促进雏鸡肝脏抗炎、抗氧化能力而发挥保护肝损伤作用,该研究同时为杜鹃素的兽医临床应用提供了试验依据和参考。
Liver injury induced liver cirrhosis,fibrosis and other pathological changes not only seriously threaten human health,but also seriously restrict the development of animal husbandry.Traditional Chinese medicine has many advantages,such as more targets,less toxic and side effects,low price,not easy to produce drug resistance,no veterinary drug residues.Farrerol is a kind of flavonoid extracted from Rhododendron.The research report shows that farrerol has antioxidant,anti-inflammatory and antibacterial activities.In this study,the chicks were pretreated with farrerol.The liver injury model was established by lipopolysaccharide(LPS)and D-galactosamine(GalN).The effect of farrerol on liver injury was determined by body weight monitoring,HE staining,enzyme activity detection and qRT-PCR.Histopathology showed that the model group had obvious inflammatory cell infiltration and disorder of hepatocyte cords.The results showed that LPS/GalN significantly increased the enzyme activity of ALT and AST in serum.The results showed that farrerol significantly decreased SOD and GSH activity,and IL-1β,TNF-α,iNOS and COX2 expression level in LPS/GalN treated.In conclusion,the liver tissue structure and liver function indexes after pretreatment with farrerol showed good preventive and therapeutic effects in the experimental period,which provided experimental basis and reference for the veterinary clinical application of farrerol.
