摘要
目的探究卵巢癌组织锌指E盒同源结合蛋白1(ZEB1)、微小RNA-574-3p(miR-574-3p)表达水平与患者临床病理特征及预后的关系。方法选取2015年1月至2018年1月于浙江医院行手术治疗的90例卵巢癌患者的癌组织作为卵巢癌组,选取其相应癌旁正常组织作为对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)法检测两组组织中miR-574-3p、ZEB1mRNA的表达水平;采用Kaplan-Meier法分析卵巢癌组织中miR-574-3p、ZEB1的表达与患者生存时间的关系;采用多因素Cox回归分析影响卵巢癌患者预后的相关因素。结果与对照组比较,卵巢癌组组织中miR-574-3p表达水平较低(P<0.05),ZEB1表达水平较高(P<0.05);卵巢癌组组织中miR-574-3p与ZEB1mRNA的表达呈负相关(P<0.05);miR-574-3p、ZEB1均与患者淋巴结或腹膜转移、国际妇产科联盟(FIGO)分期相关(P<0.05),ZEB1低表达的卵巢癌患者平均生存时间显著长于ZEB1高表达的卵巢癌患者(P<0.001),miR-574-3p低表达的卵巢癌患者平均生存时间显著短于miR-574-3p高表达的卵巢癌患者(P<0.001);miR-574-3p低表达、ZEB1高表达是影响卵巢癌患者不良预后的独立危险因素(P<0.05)。结论卵巢癌组织中ZEB1表达上调,miR-574-3p表达下调,且其与患者淋巴结或腹膜转移、FIGO分期相关,可能成为卵巢癌治疗的新靶点。
Objective To analyze the expression levels of zinc finger E-box-binding homeobox 1(ZEB1) and microRNA-574-3 p(miR-574-3 p) in patients with ovarian cancer, and to explore their relationship with the clinicopathological characteristics and prognosis of patients. Methods The cancer tissues of 90 patients with ovarian cancer who were surgically treated in Zhejiang Hospital from January 2015 to January 2018 were selected as the ovarian cancer group, and the corresponding adjacent normal tissues were selected as the control group. The expression levels of miR-574-3 p and ZEB1 mRNA in all tissues were detected with real-time fluorescence quantitative polymerase chain reaction(qRT-PCR). The relationship between the expression of miR-574-3 p and ZEB1 in ovarian cancer tissues and the survival rate of patients was analyzed with Kaplan-Meier method;the factors affecting the prognosis of ovarian cancer were analyzed with multivariate Cox regression analysis. Results Compared with the control group, the relative expression level of miR-574-3 p in the ovarian cancer group was lower(P<0.05), and the relative expression level of ZEB1 was higher(P<0.05);the expression levels of miR-574-3 p and ZEB1 mRNA in cancer tissues of patients with ovarian cancer were negatively correlated(P<0.05);both miR-574-3 p and ZEB1 were related to lymph node or peritoneal metastasis and the International Federation of Gynecology and Obstetrics(FIGO) staging(P<0.05), the average survival time of ovarian cancer patients with low ZEB1 expression was significantly longer than that of ovarian cancer patients with high ZEB1 expression(P<0.001), the average survival time of ovarian cancer patients with low miR-574-3 p expression was significantly shorter than that of ovarian cancer patients with high miR-574-3 p expression(P<0.001);the low expression of miR-574-3 p and the high expression of ZEB1 were independent risk factors affecting the poor prognosis of patients with ovarian cancer(P<0.05). Conclusions The expression of ZEB1 is up-regulated and the expression of miR-574-3 p is down-regulated in ovarian cancer tissues, which are related to ovarian cancer lymph node or peritoneal metastasis, FIGO staging, and may become new therapeutic targets for ovarian cancer.
