Toll样受体2、Toll样受体4在梅毒血清固定患者中的表达及与梅毒细胞免疫的相关性研究

Expression of TLR2 and TLR4 in serum-fixed syphilis patients and their relationship with syphilis cell immunity

目的分析Toll样受体(TLR)2、TLR4在梅毒血清固定患者中的表达及与梅毒细胞免疫的相关性。方法选取2019年3月至2021年8月峨眉山市中医医院诊治的199例梅毒患者作为研究对象,将其中未接受驱梅治疗的46例患者作为梅毒组,将驱梅治疗后血清固定的67例患者作为固定组,将驱梅治疗后血清转阴的86例患者作为转阴组;另选取50例健康体检者作为健康组。对比四组外周血单核细胞(PBMCs)中TLR2、TLR4 mRNA相对表达量及血清辅助性T细胞(Th)1/Th2型免疫应答、T淋巴细胞亚群水平,经Pearson相关性检验分析TLR2、TLR4 mRNA相对表达与Th1/Th2型免疫应答、T淋巴细胞亚群的相关性。结果PBMCs中TLR2、TLR4 mRNA相对表达量:固定组<健康组<转阴组<梅毒组(P<0.05);血清白介素-2(IL-2)、干扰素-γ(TFN-γ)、CD4^(+):固定组<梅毒组<转阴组<健康组,IL-4、IL-10、CD8^(+):固定组>梅毒组>转阴组>健康组(P<0.05);TLR2、TLR4mRNA相对表达与IL-2、TFN-γ、CD4^(+)呈正相关(r>0,P<0.05),与IL-4、IL-10、CD8^(+)呈负相关(r<0,P<0.05)。结论梅毒血清固定患者PBMCs中TLR2、TLR4呈低表达,二者表达与Th1/Th2型免疫应答、T淋巴细胞亚群水平密切相关。

Objective To analyze the expression of Toll-like receptor2(TLR2)and Toll-like receptor 4(TLR4)in syphilis in serum-fixed patients and their relationship with syphilis cell immunity.M ethods A total of 199 patients with syphilis diagnosed and treated in Emeishan Hospital of Traditional Chinese Medicine from March 2019 to August 2021 were selected as the research subjects.Among them,46 patients who were not treated with Qumei therapy were taken as the sy philis group,67 patients with serum fixation after Qumei therapy were taken as the fixation group,and 86 patients whose serum became negative after Qumei therapy were taken as the negative conversion group.Another 50 healthy people were selected as the health group.The relative expression levels of TLR2 and TLR4 mRNA in peripheral blood mononuclear cells(PBMCs)of the four groups were compared,as well as the levels of helper T cell(Th)1/Th2 immune response and T lymphocyte subsets in serum.The correlation between TLR2 and TLR4 mRNA relative expression and Th1/Th2 immune response and T lymphocyte subsets was analyzed by Pearson correlation analysis.Results Relative expression levels of TLR2 and TLR4 mRNA in PBMCs:fixation group<health group<negative conversion group<syphilis group(P<0.05);Serum interleukin(IL)-2,interferon(TFN)-γ,CD4^(+):fixation group<syphilis group.<negative conversion group<health group;IL4,IL-10,CD8^(+):fixation group>syphilis group>negative conversion group>health group(P<0.05).Relative expression of TLR2 and TLR4 mRNA was positively associated with IL-2,TFN-γ,and CD4^(+)(r>0,P<0.05),and was negatively associated with IL4,IL-10 and CD8^(+)(r<0,P<0.05).Conclusions TLR2 and TLR4 were low expressed in PBMCs of syphilis serum fixation patients,and their expression was closely associated with Th1/Th2 immune response and T lymphocyte subset level.
