红景天沙棘口服液对酒精致小鼠急性肝损伤的保护作用检测

Protective effects of Rhodiola Rosea-Hippophae Rhamnoides Drink on alcohol-induced liver damage in mice

目的 探讨红景天沙棘口服液对急性酒精性肝损伤的保护功能。方法 体质量18 g~22 g的雄性KM小鼠60只,随机分为空白对照、模型对照和红景天沙棘口服液2.5 ml/kg、5.0 ml/kg、15.0 ml/kg等5个组,连续灌胃30 d。用酒精灌胃造成酒精性肝损伤模型。测定肝组织中MDA、GSH含量,TG的含量和蛋白质含量,检测小鼠肝组织脂肪改变程度。结果 红景天沙棘口服液5.0 ml/kg、15.0 ml/kg组小鼠肝脏GSH含量分别为(1.45±0.26) mg/g和(1.67±0.24) mg/g,TG含量分别为(15.2±3.0)μmol/g和(13.4±3.7)μmol/g,红景天沙棘口服液15.0 ml/kg组小鼠肝脏MDA含量为(0.69±0.17) nmol/mgprot,与模型对照组相比差异有统计学意义(P <0.05)。红景天沙棘口服液5.0 ml/kg、15.0 ml/kg组的肝脏脂肪变性明显低于模型对照组。结论 红景天沙棘口服液可减轻酒精引起的小鼠肝损伤,对酒精性肝损伤有保护作用。

Objective To explore the protective effects of Rhodiola Rosea-Hippophae Rhamnoides Drink on alcohol-induced liver damage in mice. Methods Sixty male KM mice were divided into following groups randomly: blank control group,model control group with liver damage and 3 experimental groups in which the doses of Rhodiola Rosea-Hippophae Rhamnoides Drink were 2. 5 ml/kg,5. 0 ml/kg and 15. 0 ml/kg,respectively,and continuous gavage was conducted for 30 days. Alcoholic liver injury model was induced by alcohol intragastric administration. The contents of malondialdehyde( MDA) and glutathione( GSH) in liver tissue were determined by colorimetry. The content of triglyceride( TG) and protein in liver tissue were measured by biochemistry analyzer. The pathological change of fat in liver tissue was measured by automatic frozen microtome.Results The contents of GSH were( 1. 45 ± 0. 26) mg/g and( 1. 67 ± 0. 24) mg/g in 5. 0 ml/kg and 15. 0 ml/kg groups. The contents of TG were( 15. 2 ± 3. 0) μmol/g and( 13. 4 ± 3. 7) μmol/g in 5. 0 ml/kg and 15. 0 ml/kg groups. MDA content in15. 0 ml/kg group was( 0. 69 ± 0. 17) nmol/mgprot,which was significantly different from that in model control group( P <0. 05). The pathological changes of hepatic steatosis in 5. 0 ml/kg and 15. 0 ml/kg groups were significantly better than that in model control group. Conclusion Rhodiola Rosea-Hippophae Rhamnoides Drink can reduce the liver injury induced by alcohol in mice and protect the liver from alcohol-induced injury.
