佩兰黄酮的纯化及其抗氧化与抗运动疲劳作用 被引量:7

Purification of Flavonoids from Eupatorium fortunei Turcz.and Its Anti-oxidant and Resisting Exercise Fatigue Effects

摘要 研究大孔树脂纯化佩兰黄酮提取物的最佳条件及其抗氧化与抗运动疲劳活性。采用单因素实验确定该树脂纯化佩兰黄酮提取物的最佳条件,并通过体外抗氧化实验和动物实验研究不同产物的抗氧化与抗运动疲劳活性。结果表明,最佳纯化条件:上样浓度与流速分别为3.0 mg/L和1.0 mL/min,上样液pH与体积分别为4.0与60 mL,洗脱液乙醇体积分数与体积分别为70%与100 mL,洗脱流速为1.0 mL/min,产物的黄酮纯度由24.2%增大至69.4%。纯化后的佩兰黄酮对羟基自由基(·OH)与超氧阴离子自由基(O_(2)^(-)·)的半抑制浓度(IC_(50))分别为0.47、0.42 mg/mL,抗氧化能力增强,可明显延长小鼠的运动时间,降低其运动后乳酸与尿素氮浓度,并有助于增强SOD与GSH-Px的活力,从而减少丙二醛的生成,因此具有较好的抗运动疲劳活性,可为佩兰黄酮利用提供参考。 This work investigated the condition of macroporous resin purified of flavonoids extract from Eupatorium fortunei Turcz.and effects of anti-oxidation and resisting exercise fatigue.The optimum purification condition was confirmed by single factor experiment,meanwhile the activity of anti-oxidation and resisting exercise fatigue of different product were researched by free radical scavenging experiment and animal experiment,respectively.The results showed that the optimum purification conditions were as follows:The sample concentration was 3.0 mg/L,loading flow rate was1.0 mL/min,the pH of sample solution was 4.0,and the volume of sample solution was 60 mL,and the 100 mL of 70%ethanol solution with 1.0 mL/min of desorption velocity.The purity of flavonoids in product increased from 24.2%to69.4%.The anti-oxidation capacity of purified product was higher than crude extract,and the IC_(50)values for·OH and O_(2)^(-)·were 0.47 and 0.42 mg/mL,respectively.Compared with blank control group and crude extract group,the purified product could evident prolonged exercise time of mice,decreased concentrations of lactic acid and blood urea nitrogen after exercise of mice,and possessed enhancement activities of SOD and GSH-Px,then decreased the concentration of malondialdehyde in vivo.Therefore,it had better resisting exercise fatigue effect,and the research could provide a reference for exploit of Eupatorium fortunei Turcz.
