摘要
目的:Box-Behnken设计-响应面法优选香青兰中挥发油的最佳提取工艺。方法:采用Box-Behnken响应面法,以柠檬醛、乙酸香叶酯、香叶醇含量和挥发油量的总评归一值为评价指标,考察提取加水量、浸泡时间和提取时间对香青兰挥发油提取工艺的影响。结果:挥发油的提取工艺最优值为:加水量3.059倍,浸泡时间15.648 h,提取时间5.097 h,预测值OD=0.865;根据实际生产选择优化条件为:加水量3倍,浸泡时间16 h,提取时间5 h,通过3批次验证实验,OD分别为0.859 7,0.874 2,0.852 1,RSD=1.30%。结论:优选得到香青兰挥发油的提取工艺稳定可靠,重复性好。
Objective: To optimize the extraction process of volatile oil from Dracocephalum moldavica L. by Box-Behnken design response surface methodology(BBD-RSM). Methods: Based on BBD-RSM, the contents of citral, geraniyl acetate, geraniol and essential oil were used as the evaluation indexes to investigate the effects of water addition amount, soaking time and extraction time on the extraction process of essential oil from Dracocephalum moldavica L.. Results: The optimal extraction conditions were as follows: adding 3.059 times of water, soaking for 15.648 h and extracting for 5.098 h, and the predicted value OD=0.865. According to the actual production, the optimal conditions were selected as 3-fold amount of water added, soaking for 16 hours and extracting 5 hours, and the OD of three batches of parallel samples was 0.859 7, 0.874 2 and 0.852 1, respectively with the RSD of 1.30%. Conclusion: The optimized extraction process of volatile oil from Dracocephalum moldavica L. is proved to be stable and reliable with high repeatability.
