重组植酸酶蛹虫草的制备及其对肉仔鸡胫骨发育的影响 被引量:1

Preparation of Recombinant Phytase Crdyceps militaris and Its Effect on Tibia Development of Broilers

摘要 为了研究重组植酸酶蛹虫草对艾维因肉鸡生长性能、胫骨发育和免疫器官指数的影响,试验利用PEG转化法将已构建成功的真菌表达载体pCB130NG-phyA转化蛹虫草原生质体,成功获得重组植酸酶基因蛹虫草菌丝体。将其冻干粉作为饲料添加剂,以144只肉仔鸡为研究对象,随机分6组,每组3次重复,每次重复8只。结果表明:重组蛹虫草中植酸酶蛋白的表达量最高约占总可溶蛋白的(0.091±0.006)%,酶活力为402.2 U/g。在21日龄和42日龄,重组植酸酶剂量500 U/kg和1000 U/kg组对肉鸡胫骨中粗蛋白含量、钙含量和磷含量比对照组显著提高(P<0.05)。说明日粮添加重组植酸酶蛹虫草可显著促进矿物元素在肉仔鸡胫骨的沉积。 In order to study the effects of recombinant phytase Cordyceps militaris on growth performance,tibia development,apparent digestibility and immune organ index of avian broilers,the constructed fungal expression vector pCB130NG-phyA was transformed into pupae grassland protoplasts by PEG transformation method,then the mycelium of Cordyceps militaris with recombinant phytase gene was successfully obtained.144 one-day-old AA broilers were randomly divided into six treatment groups,each with 3 replicates,and each replicate had 8 broilers.The results show that the highest expression of phytase protein in recombinant Cordyceps militaris was about(0.091±0.006)%of total soluble protein,and enzyme activity was 402.2 U/g.At the age of 21 and42,ash content,phosphorus content and calcium content of tibial index of broilers treated with recombinant phytase dosage 500 U/kg and 1000 U/kg were significantly improved compared with those of the control group(P<0.05),indicating that adding recombinant phytase Cordyceps militaris diet can significantly promote the deposition of mineral elements in the tibia of AA broilers.
