摘要
本研究探讨天麻钩藤加味饮(TGJY)对肝阳上亢型高血压大鼠的治疗作用。采用N’-硝基-L-精氨酸(L-NNA)和附子汤诱导制备肝阳上亢型高血压大鼠模型。将36只大鼠随机分成6组,空白组、模型组、卡托普利组、TGJY低剂量组、TGJY中剂量组和TGJY高剂量组,每组6只。空白对照组、模型组给予去离子水灌胃,卡托普利组给予卡托普利灌胃,TGJY低、中、高剂量组给予不同浓度的天麻钩藤加味饮灌胃。分别在造模前后以及给药2周后测量大鼠的饮水量、血压和检测易怒程度,末次给药后记录各组大鼠肾脏系数、主动脉形态以及内皮素-1(ET-1)和一氧化氮(NO)含量。结果显示,相对于正常组,模型组饮水量、血压、易怒程度和ET-1含量显著升高,NO含量显著下降(P<0.01),主动脉形态受损。给药治疗后与模型组相比,大鼠各项指标都得到不同程度的改善。尤其是TGJY高剂量组,与模型组相比饮水量、血压、易怒程度、ET-1和NO水平具有显著性改善(P<0.05),与卡托普利组相比易怒程度也具有显著性下降(P<0.05)。天麻钩藤加味饮可以通过降低高血压大鼠饮水量、血压、易怒程度、肾脏系数和ET-1含量,增加NO含量,改善主动脉形态受损,从而对肝阳上亢型高血压大鼠起到治疗效果。
The present study investigated the therapeutic effect of Tianma Gouteng Jiawei decoction(TGJY) on hyperactive liver-yang hypertensive rats. N’-nitro-L-arginine(L-NNA) and Fuzi soup were used to induce the preparation of a hyperactive liver-yang hypertension rat model. The 36 rats were randomly divided into 6 groups, nomal group, model group, captopril group, TGJY low-dose group, TGJY mediumdose group and TGJY high-dose group, 6 rats in each group. The blank control group and model group were given deionized water by gavage,the captopril group was given captopril by gavage, and the low,medium and high dose groups were given different concentrations of Tianma Gouteng Jiawei decoction by gavage. Water consumption, blood pressure, and irritability were measured before and after modeling and two weeks after drug administration. The renal coefficients, aortic morphology and nitric endothelin-1(ET-1) and Nitric oxide(NO) contents were recorded in each group after the last administration. The results show that, relative to the normal group, the model group had significantly higher drinking water, blood pressure, irritability, kidney coefficient, ET-1 content, significantly lower NO content(P<0.01), and impaired aortic morphology. After the administration of the drug, all the indicators of the rats were improved to different degrees compared with the model group. In particular, the high-dose group showed significant improvement in drinking water, blood pressure, irritability, ET-1 and NO levels compared with the model group(P<0.05), and a significant decrease in irritability compared with the captopril group(P<0.05). Tianma Gouteng Jiawei decoction can have a therapeutic effect on hypertensive rats with hyperactive liver-yang hypertension by reducing drinking water, blood pressure, irritability, renal coefficient, ET-1 content, increasing NO content, and improving the impaired aortic morphology.
