HPLC法测定沙棘雨生红球藻胶囊中异鼠李素的含量 被引量:1

摘要 本研究采用高效液相色谱法(HPLC)建立了沙棘雨生红球藻胶囊中异鼠李素的含量测定方法。沙棘雨生红球藻胶囊内容物经95%乙醇提取后进行酸处理,使苷元充分水解为苷,采用InertSustainSwift-C(4.6 mm 250 mm,5 m)柱作为固定相;甲醇-0.4%磷酸溶液(56:44)作为流动相;流速为1.0 mL/min;检测波长为370 nm;柱温25℃;进样体积10 L。结果显示异鼠李素在0.004 38~0.011 68 mg/mL的质量浓度范围内线性关系良好(R=0.999 4),回收率在96.43%~98.98%范围内,RSD=0.89%(n=6)。该方法测得3批产品中异鼠李素的含量均值为45.26 mg/100 g(n=9)。该测定方法准确高效、简单易行,可为沙棘雨生红球藻胶囊中异鼠李素的含量测定提供参考。 In this study, a high performance liquid chromatographic(HPLC) method was established for the determination of isorhamnetin in Shaji Haematococcus pluvialis capsules. The contents of Shaji Haematococcus pluvialis capsules were extracted with 95% ethanol and treated with acid to hydrolyze aglylate into glycosides. Inert Sustain Swift Ccolumn(4.6 mm 250 mm, 5 m) was used as the stationary phase;Methanol-0.4% phosphoric acid aqueous solution(56:44) was used as mobile phase;The flow velocity was 1.0 mL/min;The detection of UV wavelength was 370 nm;The temperature of column was 25 ℃ and the injection volume was 10 L. The results showed the linearity of isorhamnetin was good in the mass concentration range of 0.004 38 ~0.011 68 mg/mL(R~2=0.999 4), and the recoveries were in the range of 96.43% to 98.98% with RSD=0.89%(n=6). The mean value of isorhamnetin in three batches of products measured by this method was 45.26 mg/100 g(n=9). This determination method is accurate, efficient and easy to perform, which would provide a reference for the establishment of quality standards for isorhamnetin in Shaji Haematococcus pluvialis capsules.
