副溶血性弧菌能力验证鉴定分析 被引量:2

Identification and Validation of Vibrio Parahaemolyticus

摘要 为确保能够维持较高的检测水平,出具的检测结果具有准确性和有效性,本实验室参加食品中副溶血性弧菌(定性)检测能力验证,监督和提升实验室检测能力。此次能力验证样品为西林瓶真空包装的白色冻干块状样品两瓶(样品编号分别为S073和R813),依据国家标准GB 4789.7-2013,采取手工生化试验、生化鉴定套装和全自动微生物生化鉴定系统对能力验证样品进行鉴定,并且采用荧光PCR对其结果进行确认。结果显示:样品S073每25 mL中未检出副溶血性弧菌;样品R813每25 mL中检出副溶血性弧菌,分离出的菌株生化鉴定结果中手工生化嗜盐性试验结果不典型,其余结果都符合副溶血性弧菌标准结果,全自动微生物生化鉴定系统结果符合率较高,荧光PCR结果呈阳性,因此不能仅凭个别生化试验不明显就判定为阴性结果,需要结合多种方法进行最终确认。此次能力验证结果得到满意评价,为以后实验室食品中该项目的检验提供了指导和依据。 In order to ensure that the laboratory can maintain a high level of testing, and its test results issued are accurate and effective, participate in the "Verification of Vibrio Parahaemolyticus(Qualitative) Testing Ability in Food" to supervise and improve the laboratory testing ability. The samples for this capability verification are two vacuum-packed white freeze-dried block samples(sample numbers are S073 and R813 respectively). According to the national standard GB 4789.7-2013, manual biochemical test,biochemical identification kit and automatic microbial biochemical identification system were used to identify the ability verification samples, and fluorescence PCR was used to confirm the results. The results showed that no Vibrio Parahaemolyticus was detected in sample S073 per 25 mL. Vibrio Parahaemolyticus was detected in sample R813 every 25 mL. The biochemical identification results of the isolated strains were not typical in the manual biochemical halophilic test, and the other results were consistent with the standard results of Vibrio parahaemolyticus. The results of the automatic microbial biochemical identification system had a high coincidence rate, and the fluorescence PCR results were positive. Therefore, a negative result cannot be determined only by the insignificance of individual biochemical tests, and it needs to be confirmed by a variety of methods. The results of the competency verification received satisfactory evaluation, which provided guidance and basis for the future food testing of the project in laboratory.
