2型糖尿病患者血清GPR120水平与脂质代谢的相关性 被引量:2

Correlation between serum GPR120 level and lipid metabolism in patients with type 2 diabetes mellitus

摘要 目的探讨2型糖尿病(T2DM)患者血清G蛋白偶联受体120(GPR120)表达水平及其与脂质代谢的相关性。方法收集56例T2DM患者(T2DM组)和56例健康体检者(对照组)静脉血。采用液相色谱-质谱联用方法检测血清脂质代谢产物,RT-PCR检测静脉血白细胞中GPR120和成纤维细胞生长因子21(FGF21)mRNA水平,分析脂质代谢情况与GPR120和FGF21 mRNA相对表达量的相关性。在人单核巨噬细胞THP-1中加入终浓度分别为0、0.01、0.1、1、10和20μmol/L的GPR120激动剂TUG891刺激6 h后,采用RT-PCR检测THP-1细胞中GPR120和FGF21 mRNA表达水平。结果与对照组相比,T2DM组血清磷脂酸、胆固醇酯、二酰甘油、卵磷脂等表达量上调,鞘氨醇和磷脂酰乙醇胺表达量下调(P<0.01),白细胞中GPR120 mRNA[3.43(2.00,61.27)vs.0.99(0.56,1.73)]和FGF21 mRNA[2.97(1.91,5.12)vs.0.99(0.66,1.51)]相对表达量升高(P<0.01)。T2DM组GPR120及FGF21 mRNA相对表达量与差异性脂质的表达量呈正相关(P<0.05)。与0μmol/L相比,0.1μmol/L及以上浓度的TUG891刺激THP-1细胞后GPR120和FGF21 mRNA相对表达量增加(P<0.01)。结论T2MD患者脂质代谢紊乱,GPR120表达水平升高且与差异性脂质的表达量呈正相关。GPR120通过FGF21对T2DM脂质代谢紊乱进行调控。 Objective To explore the correlation between serum level of G-protein-coupled receptor 120(GPR120)and lipid metabolism in the patients with type 2 diabetes mellitus(T2DM).Methods The venous bloods of 56 T2DM patients(group A)and 56 healthy people(group C)were collected.Lipid profiles were determined by liquid chromatography-mass spectrometry and the expression levels of GPR120 and fibroblast growth factor 21(FGF21)mRNA in leukocytes were measured by RT-PCR.The correlations of lipid metabolism with the expressions of GPR120 and FGF21 mRNA were analyzed.The expression levels of GPR120 and FGF21 mRNA in THP-1 cells were measured by RT-PCR after stimulation with final concentrations of GPR120 agonist TUG891 at 0,0.01,0.1,1,10 and 20μmol/L for 6 hours,respectively.Results Compared with group C,serum expression levels of phosphatidic acid,cholesterol lipid,diglyceride,lecithin and other lipids were increased,serum expression levels of sphingosine and phosphatidylethanolamine were decreased(P<0.01),and the expressions of GPR120 mRNA[3.43(2.00,61.27)vs.0.99(0.56,1.73)]and FGF21 mRNA[2.97(1.91,5.12)vs.0.99(0.66,1.51)]in leukocytes were increased in group A(P<0.01).The expressions of GPR120 and FGF21 mRNA were positively correlated with the expression levels of differential lipids in group A(P<0.05).Compared with 0μmol/L,the expressions of GPR120 and FGF21 mRNA were increased after stimulation of THP-1 cells with TUG891 at the concentrations≥0.1μmol/L(P<0.01).Conclusion Lipid metabolism is disordered,and the expression of GPR120 is increased and positively correlated with the expression levels of differential lipids in the blood of T2 MD patients.GPR120 regulates lipid metabolism disorder through FGF21 in T2DM.
