摘要
目的观察中药单体芍药苷及丹皮酚对hERG、Iks、Katp、Cav 3.2、HCN_(2)、HCN4等离子通道的作用,探讨2种中药单体在离子通道水平上的作用机制。方法运用全细胞膜片钳技术观察1 uM及10 uM 2种不同浓度芍药苷及丹皮酚对hERG、Cav 3.2、Iks、Katp、HCN2、HCN4等离子通道电流的抑制作用。结果2种浓度的芍药苷及丹皮酚均对离子通道的电流具有一定的抑制作用,10 uM浓度药物对离子通道电流的阻断作用普遍优于1 uM。针对不同的离子通道,芍药苷对Cav 3.2离子通道的抑制作用较明显,丹皮酚对IKs通道的抑制作用较显著。结论2种中药单体对多种离子通道均有一定的阻断作用,且呈浓度依赖性,这可能是2种中药单体发挥抗心律失常的作用机制之一。
Objective To observe the effects of traditional Chinese medicine monomers paeoniflorin and paeonol on hERG,Iks,KTP,Cav3.2,HCN_(2),HCN4 and other ion channels,and to explore the mechanism of the two Chinese medicine monomers at the level of ion channels.Methods The whole-cell patch clamp technique was used to observe the inhibitory effects of paeoniflorin and paeonol at two different concentrations of 1 uM and 10 uM on hERG,CaV3.2,Iks,Katp,HCN2,HCN4 and other ion channel currents.Results Paeoniflorin and paeonol at the two concentrations had certain inhibitory effects on the currents of ion channels,and the blocking effect of the two drugs at 10uM on the currents of ion channels was generally better than that at 1uM.For different ion channels,paeoniflorin had a more obvious inhibitory effect on the Cav3.2 ion channel,while paeonol had a more significant inhibitory effect on the IKs channel.Conclusion The two Chinese medicine monomers have certain blocking effects on a variety of ion channels in a concentration-dependent manner,which may be one of the antiarrhythmic mechanisms of the two Chinese medicine monomers.
