Optimization of extraction pigment from Chinese wild rice by response surface methodology

摘要 以中国菰米为原料,采用响应面法优化其色素提取工艺。从液料比、温度、时间和乙醇中的盐酸浓度4个单因素试验中得出最佳参数,通过响应面试验研究分析液料比、时间和乙醇中的盐酸浓度对中国菰米色素提取的影响。结果表明,二次回归模型对实验结果有较好的拟合效果。液料比和盐酸浓度对色素提取有极显著影响,各因素之间交互作用的影响显著。温度70℃、液料比4.5mL/g、时间182min、0.85%盐酸浓度的乙醇溶液为最优提取工艺参数。此条件下,色素的吸光度值可达0.748。 Using Chinese wild rice as raw material,the response surface method was used to optimize the extraction of pigment.Firstly,liquid-solid ratio,temperature,time and the concentration of hydrochloric acid in ethanol were tested by single factor experiment,and then the optimal parameters were obtained.Then,the effects of liquid-solid ratio,time and concentration of hydrochloric acid in ethanol on the extraction of pigment from Chinese wild rice were studied and analyzed by response surface experiment.The experimental results showed that the quadratic regression model had a good fitting effect on the experimental results,liquid-solid ratio and hydrochloric acid concentration had extremely significant effects on pigment extraction,and the interaction between these factors was significant.The optimum extraction conditions were as follows:temperature 70℃,ratio of liquid to material 4.5mL/g,time 182min,and ethanol solution with 0.85%hydrochloric acid concentration.Under this condition,the absorbance value of pigment could reach 0.748.
