摘要
目的:建立了高效液相色谱法测定婴幼儿配方乳粉和保健品中β-胡萝卜素的方法。方法:通过对GB 5009.83—2016测定食品中胡萝卜素前处理方法的优化,优选婴幼儿配方乳粉和保健品中β-胡萝卜素的适配性前处理方法,采用Agela C_(18)色谱柱(250mm×2.1mm,5μm)进行分离,以甲醇∶乙腈∶三氯甲烷为流动相,梯度洗脱,基质标准曲线定量。结果:β-胡萝卜素在0.6~120μg/mL范围内线性关系良好,GB 5009.83—2016皂化-萃取法的回收率为70.2%~103.9%,相对偏差均小于8.3%(n=6);采用本研究溶剂萃取法前处理的回收率为75.7%~98.7%,相对偏差为均小于7.8%(n=6)。结论:本实验建立的方法前处理简单,样品损失率低,结果准确可靠,适合婴幼儿配方乳粉和保健品中β-胡萝卜素的测定。
Objective:A method of high performance liquid chromatography(HPLC)was developed to determineβ-carotene in infant formula and dietary supplements.Methods:A suitable pretreatment method ofβ-carotene in infant formula and dietary supplements was optimized by optimizing the pretreatment method and comparing the pretreatment method with GB 5009.83-2016 National standard for food safety-Determination of carotenein food,the analytes were separated on an Agela C_(18)(250mm×2.1mm,5μm)column,the mobile phase was methanolacetonitrile-chloroform.β-carotene was quantified by matrix standard curve.Results:The results showed a good linear relationship within the range of 0.6~120μg/mLβ-carotene,the average recoveries of the pretreatment method(GB 5009.83—2016)were 70.2%~103.9%,with relative standard deviations(RSDs)lower than 8.3%(n=6).The average recoveries with the solvent extraction method established in this study were 75.7%~98.7%with the relative standard deviations(RSDs)lower than 7.8%(n=6).Conclusion:The established method is simple,accurate and selective,and is suitable for the determination ofβ-carotene in infant food and dietary supplements.
