摘要
目的 探讨骨髓间充质干细胞外泌体对人结直肠癌细胞系HCT116增殖、侵袭的影响及可能机制。方法 细胞分为对照组、模型组和外泌体处理组。采用CCK-8法和克隆形成实验测定细胞增殖;采用细胞划痕实验和Transwell实验检测肿瘤细胞迁移能力;采用Western blot技术检测细胞ERK2和MAPK蛋白表达。结果骨髓间充质干细胞外泌体可以有效抑制细胞增殖,且随着浓度的增加,抑制效果增强。外泌体干预显著降低人结直肠癌细胞系HCT116的迁移能力,降低人结直肠癌细胞系HCT116中ERK2和MAPK蛋白表达(P<0.05)。结论骨髓间充质干细胞外泌体能够抑制人结直肠癌细胞系HCT116增殖和迁移能力,其作用机制可能与调控ERK/MAPK通路有关。
Objective To investigate the effect of bone marrow mesenchymal stem cell exosomes on the proliferation and invasion of human colorectal cancer cell line HCT116, and to explore its possible mechanism. Methods Cells was divided into control group, model group and exosome treatment group. Cell proliferation was determined by CCK-8test and clone formation assay. Cell scratch assay and Transwell assay were used to detect the migration of tumor cells. The expression of ERK2 and MAPK was detected by Western blot. Results Bone marrow mesenchymal stem cell exosomes can effectively inhibit cell proliferation, and the inhibitory effect is enhanced with the increase of concentration. Exosome intervention significantly reduced the migration ability of human colorectal cancer cell line HCT116 and the expression of ERK2 and MAPK(P<0.05). Conclusion Bone marrow mesenchymal stem cell exosomes can inhibit the proliferation and migration of human colorectal cancer cell line HCT116, and the mechanism may be related to ERK/MAPK pathway.
