正交试验法与Box-Behnken响应面法优选白残花-乌梅药对提取工艺比较

Comparison of the Orthogonal Test and the BBD-RSM in Optimizing the Extraction Process of Rosae Multiflorae Flos-Mume Fructus Drug Pair

目的优选白残花-乌梅药对的最佳提取工艺。方法以加水量、提取时间、提取次数为考察因素,以金丝桃苷含量及浸膏得率所得综合评分为评价指标,分别采用L(934)正交试验法和Box-Behnken响应面法优选白残花-乌梅药对的提取工艺,并对2种方法的试验结果进行比较,确定最适合的提取方法。结果正交试验法优选的提取工艺为,加20倍量水,提取3次,每次1.0 h,综合评分为90.28分;Box-Behnken响应面法优选的提取工艺为,加25倍量水,提取3次,每次1.0 h,综合评分为97.85分。结论相较于正交试验法,Box-Behnken响应面法优选的提取工艺更适合提取白残花-乌梅药对。

Objective To optimize the extraction process of Rosae Multiflorae Flos-Mume Fructus drug pair.Methods Taking the amount of water addition,extraction time and extraction times as the investigation factors,and taking the comprehensive score of hyperoside content and extract yield as the evaluation index,the extraction process of Rosae Multiflorae Flos-Mume Fructus drug pair was optimized by the L9(34)orthogonal test and the Box-Behnken design(BBD)-response surface methodology(RSM)respectively.The test results of the above two methods were compared to determine the optimal extraction method.Results The optimal extraction process of the orthogonal test was as follows:adding 20 times the amount of water,extracting three times,with 1.0 h each time,and the comprehensive score was 90.28 points.The optimal extraction process of the BBD-RSM was as follows:adding 25 times the amount of water,extracting three times,with 1.0 h each time,and the comprehensive score was 97.85 points.Conclusion Compared with the orthogonal test,the BBD-RSM is more suitable for the extraction of Rosae Multiflorae Flos-Mume Fructus drug pair.
