Determination of Atractylodin and Atractyloside in Atractylodes lancea by UPLC-MS/MS

摘要 建立了茅苍术中苍术素和苍术苷超高效液相色谱—串联质谱法(UPLC-MS/MS)检测方法。通过质谱特征扫描,2种化合物均获得较高离子化效应离子对和最佳检测条件。研究表明,采用甲醇提取,液固比为80 mL/g,超声提取30 min,苍术素和苍术苷的提取量均达到较高水平,分别约为1.6 mg/g和18μg/g。该方法前处理简单、分析快速、定性定量准确、检测限低,可以满足茅苍术中苍术素和苍术苷的检测要求。 A method for determination of atractylodin and atractyloside in Atractylodes lancea by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) was developed and validated.Ion pairs and the best detection conditions with high ionization effect were obtained via feature scan of mass spectrometry.The samples were extracted with methanol at room temperature,the liquid-solid ratio was 80 mL/g,and the ultrasonic extraction time was 30 min.The extraction rates of atractylodin and atractylodin were about 1.6 mg/g and 18 μg/g.The method has the advantages of simple pretreatment,rapid analysis,accurate qualitative and quantitative analysis and low detection limit.The method can be suitable for the simultaneous analysis of Atractylodin and atractyloside in Atractylodes lancea.
