摘要
目的观察miR-124表达变化对肝癌细胞增殖,凋亡,迁移和侵袭的影响及其机制。方法检测HepG2中miR-124、ZEB2的表达变化。分别采用CCK-8、流式细胞术、Edu和Transwell技术检测miR-124与ZEB2表达变化对细胞增殖、凋亡、迁移和侵袭能力。双荧光素酶和靶基因预测miR-124与ZEB2的靶向关系;Western blot检测其对增殖、凋亡、迁移和侵袭相关蛋白表达影响。结果miR-124在HepG2细胞中的表达低于正常肝细胞L-02;而ZEB2与miR-124呈相反的趋势。生物信息学预测和双荧光素酶的结果表明,ZEB2的表达与miR-124表达负相关。与对照组相比,过表达miR-124和沉默ZEB2明显抑制细胞增殖活性、迁移和侵袭能力;沉默miR-124和过表达ZEB2明显促进细胞增殖活性、迁移和侵袭能力。Western blot结果显示,过表达miR-124和沉默ZEB2明显促进Bax蛋白表达和抑制Bcl-2、PCNA、MMP2和MMP9蛋白表达水平;沉默miR-124和过表达ZEB2则相反。结论miR-124可负向调控ZEB2抑制肝癌细胞增殖,迁移和侵袭的影响。
Aim To observe the effect of changes in miR-124 expression on the proliferation,apoptosis,migration and invasion of HCC cells and its mechanism.Methods The expression levels of miR-124 and ZEB2 were detected in HepG2 cells.CCK8,flow cytometry,Edu and Transwell were used to detect the effects of miR-124 and ZEB2 on cell proliferation,apoptosis,migration and invasion.Dual luciferase and target genes were used to predict the targeting relationship between miR-124 and ZEB2.The effect of miR-124 and ZEB2 on proliferation,apoptosis,migration and invasion-related protein expression was detected by Western blot.Results The expression of miR-124 in HepG2 cells was lower than that in normal liver cells L-02,while ZEB2 and miR-124 showed the opposite trend.The results of bioinformatics prediction and dual luciferase showed that the expression of ZEB2 was negatively correlated with the expression of miR-124.Overexpression of miR-124 and silencing ZEB2 significantly inhibited cell proliferation activity,migration and invasion ability compared with the control group;silencing miR-124 and overexpression of ZEB2 significantly promoted cell proliferation activity,migration and invasion ability.Western blot results showed that overexpression of miR-124 and silencing ZEB2 significantly promoted Bax expression and inhibited Bcl-2,PCNA,MMP2 and MMP9 expression levels.Silencing miR-124 and overexpression ZEB2 were the opposite.Conclusion miR-124 could negatively regulate the effects of ZEB2 on the proliferation,migration and invasion of HCC cells.
