甘肃地区淫羊藿指纹图谱建立及主成分分析

Fingerprint Research of Yinyanghuo(Epimedii Folium) Based on Chemical Pattern Recognition Technology

目的建立甘肃地区淫羊藿药材HPLC指纹图谱,对主要的黄酮类成分进行定量分析,结合主成分分析,评价不同产地药材质量的优劣。方法采用HPLC建立适合甘肃地区淫羊藿药材的指纹图谱。运用标准曲线法对黄酮类成分定量分析。通过SPSS软件对共有峰峰面积进行主成分分析,得出质量评价模型,评价药材质量。结果建立了甘肃地区21批淫羊藿药材HPLC指纹图谱。确定了8个共有色谱峰,指认了其中6个。相似度评价在0.497-0.980,表明甘肃地区淫羊藿药材质量差异较大。黄酮类成分定量分析结果显示,除了13号样品,其余20个样品的含量总和在1.5%之上。主成分分析提取的3个主成分累积方差贡献率达到80.064%。根据综合质量评价模型显示陇南西和县、陇南宕昌县、兰州榆中县、甘南迭部县和陇南成县等地的药材质量优于其他地方。结论建立的淫羊藿HPLC指纹图谱,方法学考察符合规定,信息丰富。结合主成分分析建立质量评价模型,为科学评价甘肃地区淫羊藿药材质量提供参考。

Objective To establish HPLC fingerprint of Yinyanghuo(Epimedii Folium)in Gansu province.The main flavonoid components were quantitatively analyzed.Combined with principal component analysis,the quality of medicinal materials from different producing areas was evaluated.Methods HPLC was used to establish the fingerprint of Yinyanghuo(Epimedii Folium)in Gansu.The standard curve method was used for quantitative analysis of flavonoids.The common peak area was analyzed by SPSS software,and the quality evaluation model was obtained to evaluate the quality of medicinal materials.Results HPLC fingerprints of 21 batches of Yinyanghuo(Epimedii Folium)samples in Gansu were established,8 common chromatographic peaks were identified and 6 of them were identified.The similarity evaluation ranged from 0.497-0.980,indicating that the quality of Yinyanghuo(Epimedii Folium)in Gansu province varied greatly.The results of quantitative analysis of flavonoids showed that except for sample NO.13,the contents of the other 20 samples were above 1.5%.The cumulative variance contribution rate of the three principal components extracted by principal component analysis reached 80.064%.The result showed that the quality of herbal medicine in Xihe county of Longnan,Min county of Longnan,Diapei county of Gannan and Hui county of Longnan were better than other places.Conclusion Fingerprint analysis combined with principal component analysis can provide reference for scientific evaluation of Yinyanghuo(Epimedii Folium)quality in Gansu province.
