基于广泛靶向代谢组学分析三个姜黄品种中姜黄素类成分 被引量:2

Analysis of curcuminoids in three Curcuma longa L.varieties based on extensive targeted metabonomics

摘要 本试验基于液相色谱-四极杆线性离子阱质谱的广泛靶向代谢组学方法,结合聚类分析和正交偏最小二乘判别分析等方法,比较分析姜黄(Curcuma longa L.)三个品种的姜黄素类代谢物的变化。结果所有样品共检出46个姜黄素类化合物,其中高含姜黄(GJH)和川姜黄(CJH)均检出46个姜黄素类代谢物,而云姜黄(YJH)检出36个。通过组间峰面积分析发现,共有41个代谢物包括指标性成分姜黄素和代表性成分去甲氧基姜黄素、双去氧基姜黄素、二氢姜黄素及四氢姜黄素的相对含量均从高到低依次为:高含姜黄、川姜黄、云姜黄。通过层次聚类和自组织映射聚类分析发现,高含姜黄与川姜黄样品可以聚为一类与云姜黄区别分开,提示高含姜黄与川姜黄质量相似而与云姜黄差异较大。结果表明,高含姜黄的代表性姜黄素类代谢物的含量高于川姜黄和云姜黄,且与川姜黄的姜黄素类代谢物组成相似,是除川姜黄外的又一高质量姜黄新品系,可为姜黄药材提供新的来源。 An extensive targeted metabonomics method based on liquid chromatography-quadrupole linear ion trap mass spectrometry combined with cluster analysis and orthogonal partial least squares discriminant analysis was used,to compare and analyze the metabolites of curcuminoids in three turmeric varieties.A total of 46 curcuminoids were detected in all samples,of which 46 curcuminoids were detected in GJH and CJH samples,while only 36 curcuminoids were detected in YJH.Analysis of the peak area between groups found that a total of 41 metabolites,including representative components curcumin,demethoxycurcumin,dideoxycurcumin,dihydrocurcumin and tetrahydrocurcumin,and their relative contents appeared as GJH>CJH>YJH.Through hierarchical clustering and self-organizing map cluster analysis,it is found that the samples of GJH and CJH can be grouped into one group and separated from YJH,suggesting that the quality of GJH is similar to CJH but is quite different from YJH.The results show that the content of representative curcumin metabolites of GJH is higher than that of CJH and YJH,and its composition of curcumin metabolites is similar to that of CJH.GJH is another high-quality new turmeric variety besides CJH,which can be used to provide new sources for turmeric medicinal materials.
