基于UHPLC-QqQ-MS对蟾衣中游离氨基酸与总氨基酸的分析 被引量:1

Analysis of free amino acids and total amino acids in toad coat based on UHPLC-QqQ-MS

摘要 为探究蟾衣的营养和药用价值,采用超高效液相色谱串联三重四级杆质谱(UHPLC-QqQ-MS)技术,分析蟾衣中游离氨基酸和总氨基酸的组成及含量。实验结果显示,在蟾衣样品中分析出的游离氨基酸有11种:天冬氨酸、谷氨酸、赖氨酸、γ-氨基丁酸、酪氨酸、苏氨酸、脯氨酸、缬氨酸、亮氨酸、异亮氨酸、苯丙氨酸;总氨基酸有18种:精氨酸、胱氨酸、组氨酸、天冬氨酸、谷氨酸、丝氨酸、赖氨酸、甘氨酸、苏氨酸、丙氨酸、γ-氨基丁酸、酪氨酸、脯氨酸、缬氨酸、蛋氨酸、亮氨酸、异亮氨酸、苯丙氨酸;并检出了17种氨基酸的含量:谷氨酸含量最高,其次是天冬氨酸,组氨酸和胱氨酸含量较低。此外,本研究建立的UHPLC-QqQ-MS技术用于测定上述游离氨基酸和总氨基酸专属性好,灵敏度高,可检测较低质量浓度的氨基酸。蟾衣的氨基酸种类丰富,药用氨基酸种类齐全,具有重要的营养价值和药用价值。实验结果可为深入研究蟾衣药材的资源开发利用提供基础资料。 In oder to explore the nutritional and medicinal value of toad coat,the composition and content of free amino acids and total amino acids in toad coat were anlyzed by ultra high performance liquid chromatography coupled with triple quadrupole mass spectrometry(UHPLC-QqQ-MS).The results showed that 11 free amino acids(aspartic acid,glutamate,lysine,γ-aminobutyric acid,tyrosine,threonine,proline,valine,leucine,isoleucine,phenylalanine)and 18 total amino acids(arginine,cystine,histidine,aspartic acid,glutamate,serine,lysine,glycine,threonine,alanine,γ-aminobutyric acid,tyrosine,proline,valine,methionine,leucine,isoleucine,phenylalanine)were detected in the toad coat.The contents of 17 amino acids were detected,and glutamate was the highest,followed by aspartic acid,while histidine and cystine were lower.In addition,UHPLC-QqQ-MS technology established in this study has good specificity and high sensitivity to determine the above free amino acids and total amino acids,and can detect amino acids with low mass concentration.There are many kinds of amino acids in toad coat,which have important nutritional and medicinal values.The results can provide basic data for the further study of the resource development and utilization of toad coat.
