益气健脾联合化疗疗法对结肠癌裸鼠p38通路调控作用的实验研究

Experimental Study on The Modulation of p38 Pathway in Nude Mice with Colon Cancer by Combining Chemotherapy With Anti-Inflammatory Mechanism of Invigorating Spleen and Replenishing Qi

目的基于益气健脾理论,使用中药抗癌复方联合希罗达,对结肠癌CT26WT细胞荷瘤裸鼠进行药物干预,通过检测各组结肠癌瘤组织中p-p38、p38蛋白的表达,深入研究益气健脾联合化疗疗法对结肠癌荷瘤裸鼠的作用机制。方法对SPF级BALB/C裸鼠进行随机分组,分别为模型组,中药抗癌复方组、希罗达组及联合用药组。裸鼠腋窝皮下注射CT26WT细胞悬液,用以制备荷瘤裸鼠模型。各组荷瘤裸鼠模型在使用药物分别干预后,测定血液常规指标,免疫蛋白印迹法(Western Blot)检测结肠癌荷瘤裸鼠瘤组织中p-p38、p38蛋白表达。结果化疗药可能影响结肠癌荷瘤裸鼠的生长,对其机体具有一定的损害性;与模型组相比较,各组中性粒细胞/淋巴细胞比值(NLR)及血小板/淋巴细胞比值(PLR)均降低,且联合用药组预后效果最为明显,抑瘤率显著高于其他组。免疫蛋白印迹检测p-p38、p38蛋白表达降低。结论益气健脾联合化疗疗法干预结肠癌,可能通过降低p-p38、p38蛋白表达抑制肿瘤生长和转移,进而抑制结肠癌疾病发展。

Objective Based on the theory of invigorating spleen and replenishing Qi,we used Chinese herbal anti-cancer compound combined with Herodah to perform drug intervention on CT26 WT cell-bearing nude mice with colon cancer.By detecting the expression of p-p38 and p38 protein in the tumor tissues of each group,the mechanism of the effect of invigorating spleen and replenishing Qi combined with chemotherapy therapy on colon cancer-bearing nude mice was investigated in depth.Methods SPF-grade BALB/C nude mice were randomLy grouped into model group,Chinese herbal anti-cancer compound group,Herodah group and combined drug group.CT26 WT cell suspension was injected subcutaneously into the axilla of nude mice to prepare a tumor-bearing nude mouse model.The expression of p-p38 and p38 protein in the tumor tissues of colon cancer nude mice was detected by immunoprotein blotting(Western Blot)after the intervention with drugs in each group.Results Chemotherapeutic drugs may affect the growth of colon cancer nude mice with certain damage to their organism;compared with the model group,the neutrophil/lymphocyte ratio(NLR)and platelet/lymphocyte ratio(PLR)were reduced in all groups,and the prognosis effect was most obvious in the combined drug group,and the tumor suppression rate was significantly higher than other groups.The expression of p-p38 and p38 protein was reduced by immunoprotein blotting.Conclusions Intervention of colon cancer with invigorating spleen and replenishing Qi combined with chemotherapy may inhibit tumor growth and metastasis by reducing p-p38 and p38 protein expression,and thus inhibit colon cancer disease progression.
