摘要
目的以蛋白质组学方法观察四君子汤对于自然衰老模型小鼠肝线粒体蛋白质变化情况,并对与其衰老存在紧密关联的蛋白标志物深入挖掘。在研究过程中,主要是就四君子汤对衰老相关蛋白质物的影响进行分析。为方证关系研究提供科学依据。方法目标研究对象筛选的是30只自然衰老雄性小鼠,空白组选择的小鼠为3月龄,其余小鼠饲养到16月龄,然后将其随机分为模型组以及四君子汤组,每组共计有10只小鼠。四君子汤组灌胃给药剂量为13 mL/kg,空白组以及模型组在进行灌胃操作上,均采用等量13 mL/kg 0.9%氯化钠溶液灌胃。取小鼠完整肝脏后从中提取线粒体样本,通过同位素标记相对和绝对定量技术,探求自然增龄模型小鼠肝细胞线粒体蛋白标志物的变化情况。结果通过对空白组以及模型组的鉴定,最终获取的差异蛋白数量是797个,而在模型组以及四君子汤组的结果分析上,最终获取的差异蛋白数量共有563个。在使用四君子汤后,回调的差异蛋白数量有83个。结论通过蛋白质组学技术研究发现,四君子汤对16月龄小鼠肝组织类固醇激素生物合成、炎症介质对TRP通道的调节、嘌呤代谢、视黄醇代谢以及ECM受体相互作用等途径均有明显的调节作用。通过上述信号通路对应蛋白的调节,四君子汤能对增龄机体功能紊乱产生相应的保护以及调节作用。
Objective To observe the effect of Sijunzi Decoction(四君子汤)on the protein changes of liver mitochondria in natural aging mice by proteomics and explore the protein markers closely related to aging.In the course of the study,we mainly analyzed the effect of Sijunzi Decoction on aging related proteins to provide a scientific basis for the study of the relationship be-tween prescription and syndrome.Methods Thirty natural aging male mice were selected as the research objects.The mice in the blank group were 3 months old,and the other mice were raised to 16 months old.Then they were randomly divided into the model group and the Sijunzi Decoction group,with 10 mice in each group.The dosage of Sijunzi Decoction group was 13 mL/kg,while the blank group and model group were given normal saline with the dosage of 13 mL/kg.The changes of mitochondrial protein markers in liver cells of natural aging mice were investigated by isotope labeling relative and absolute quantitative techniques.Re-sults Through the identification of blank group and model group,the number of differential proteins was 797,while in the analysis of the model group and the Sijunzi Decoction group,the number of differential proteins was 563.After using Sijunzi Decoction,83 differential proteins were recalled.Conclusion Through the proteomic study,Sijunzi Decoction can regulate the steroid biosynthe-sis,TRP channel regulation by inflammatory mediators,purine metabolism,retinol metabolism and ECM receptor interaction in 16-month-old mice.Through the regulation of the corresponding proteins of the above signaling pathways,Sijunzi Decoction can have the corresponding protection and regulation on the aging body dysfunction.
