基于信息熵赋权结合Box-Behnken响应面法优化北苍术提取工艺 被引量:6

Optimization of Extraction Process of Beicangzhu(Atractylodes chinensis)by Combination of Response Surface Methodology with Information Entropy Weight Method

摘要 目的利用赋权法-Box-Behnken响应面法优化北苍术的最优提取工艺。方法采用高效液相色谱法(HPLC)结合信息熵-响应面法,以白术内酯Ⅲ、白术内酯Ι、β-桉叶醇、苍术素的含量为评价指标,赋予各成分权重系数,结合Box-Behnken响应面法对提取溶剂、料液比、提取时间进行考察,计算综合评分优化北苍术提取工艺。结果优化提取工艺条件为提取溶剂甲醇浓度87.77%、超声时间44.80 min、物料比为1∶19.23(g·mL^(-1)),综合评分M为0.97。考虑到实际操作,确定最终提取工艺,即料液比为1∶20(g·mL^(-1)),提取时间为45 min、溶剂浓度为90%。通过进行3次平行验证实验,计算综合评分分别为0.9745、0.9656、0.9752,其RSD为0.55%。结论HPLC法适用于北苍术中4种成分的同时测定,优化后的提取工艺方法简便,稳定可行,可为北苍术多成分提取的工业化生产提供参考。 Objective To optimize the extraction of Beicangzhu(Atractylodes chinensis)by information entropy weight method combined with Box-Behnken response surface method.Methods High performance liquid chromatography(HPLC)combined with information entropy-response surface(RSM)was used,with the contents of atrctylenolideⅢ,atrctylenolideⅠ,atractylodin andβ-eduesmol as the evaluation indexes,each component was given the weight coefficients.Combined with the Box-Behnken response surface method,the extraction solvent,the material ratio,the extraction time and the comprehensive score were calculated to optimize the extraction technology of Beicangzhu(Atractylodes chinensis).Results The optimal extraction conditions were as follows:the extraction solvent was 87.7%,the ultrasonic time was 44.80 min,the material ratio was 1∶19.23(g·mL^(-1))and the comprehensive score was 0.97.Considering the actual operation,the final extraction process was determined.The solid-liquid ratio was 1∶20,the extraction time was 45 min and the solvent concentration was 90%.Through three parallel validation experiments,the calculated comprehensive scores were 0.9745,0.9656 and 0.9752,respectively and the RSD was 0.55%.Conclusion The HPLC method is suitable for the simultaneous determination of four components in Beicangzhu(Atractylodes chinensis),the optimized extraction method is simple,stable and feasible,and can provide a reference for the industrial production of multi component extraction of Beicangzhu(Atractylodes chinensis).
