摘要
通过体外生物技术的细胞平台,从多角度对一种水解贝壳硬蛋白进行美白方面的作用研究。结果显示水解贝壳硬蛋白Ligfactor^(TM)可以明显抑制B16小鼠黑色素瘤细胞及人原代黑素细胞的酪氨酸酶活性;在人内皮素-I刺激下,其可以显著降低人原代黑素细胞的酪氨酸活性上升的情况;同时共培养显示其可以有效抑制内皮素-1刺激下黑素细胞树突结构增多的情况;多巴染色显示加入Ligfacor^(TM)黑色素染色浅,黑素含量低,说明其具有降低黑色素合成的作用。以上结果表明,水解贝壳硬蛋白Ligfactor^(TM)有美白的功效,具有应用于美白类相关产品的潜力。
Through the in vitro cell-based platform,the effectiveness of a hydrolyzed conchiolin protein was verified from multiple levels to provide theoretical basis and scientific guidance for its application in cosmetics.Research of hydrolyzed conchiolin protein on the effects of tyrosinase activity,melanin production,and dendritic structure by using B16 mouse melanoma cells,primary human melanocytes,and keratinocyte-melanocyte co-culture systems was conducted.The effects of hydrolyzed conchitin on tyrosinase activity were also compared with or without endothelin-1 stimulation on human primary melanocytes.The results showed that this hydrolyzed conchiolin protein Ligfactor^(TM) can significantly inhibit the tyrosinase activity of B16 mouse melanoma cells and normal human epidermal melanocyte.Ligfactor^(TM) 0.1%and 0.01%inhibition rate(18.18%^(**),12.69%^(*))on B16 were higher than the positive control arbutin(11.92%^(**)).The Ligfactor^(TM) 0.01%inhibition rate(24.92%^(*))on human primary melanocytes was also higher than that of the positive control arbutin(23.04%^(*));And under the stimulation of human endothelin-1,Ligfactor^(TM) can significantly reduce the increase of tyrosine activity of human primary melanocytes,and the effect of low concentration dose of Ligfactor^(TM) is more obvious,Ligfactor^(TM)0.001%inhibition rate(36.08%^(**))is higher than Ligfactor^(TM) 0.01%(28.75%*),both better than the positive control arbutin(25.09%^(*)).Meanwhile,co-culture showed that it could effectively inhibit the increase of dendritic structure of melanocytes under stimulation by endothelin-1,and the number of dendritic structures added with Ligfactor^(TM) 0.001%under stimulation was closer to the normal level(NT without stimulated by ET-1),Dopa staining shows that the addition of Ligfactor^(TM) melanin stains lightly,and the low melanin content indicates that it has the effect of reducing melanin synthesis.The above results show that this hydrolyzed conchiolin can inhibit the activity of tyrosinase,inhibit the increase of melanin content and dendritic structure caused by ET-1 stimulation.Therefore,it has a whitening effect under normal conditions and under special stimulation(ET-1 stimulation),which provides a theoretical basis for its application in cosmetics.
