摘要
目的:为深入研究hsa-miR-20a-3p的生物学功能及相关疾病的致病机制提供一定的理论基础。方法:利用miRbase等数据库分析hsa-miR-20a-3p的物种保守性,应用Targetscan、TarBase、miRDB和miRTarBase靶基因数据库预测hsa-miR-20a-3p的靶基因,并通过DAVID数据库对靶基因进行GO功能注释和KEGG信号通路富集分析。结果:hsa-miR-20a-3p的成熟碱基序列在各物种间高度保守;预测得到156个靶基因集合;GO功能注释分析结果显示,hsa-miR-20a-3p靶基因主要富集在核浆、细胞质、细胞核等9个细胞组分,富集在蛋白质结合、染色质结合、DNA结合等11个分子功能,富集在前脑形态发生、学习或记忆、器官诱导、基因表达调控等15个生物学过程(P<0.05);KEGG信号通路显著富集在病毒致癌、乙型肝炎以及HTLV-Ⅰ感染3个信号通路(P<0.05)。结论:hsa-miR-20a-3p通过调控靶基因广泛参与多个重要的生物学过程和信号通路,尤其是病毒致癌过程是值得进一步研究的方向。
Objective:To provide a theoretical basis for the further study of the biological functions of hsa-miR-20a-3p and the pathogenesis of related diseases.Methods:Use miRbase and other databases to analyze the species conservation of hsa-miR-20a-3p,use Targetscan,TarBase,miRDB and miRTarBase target gene databases to predict the target genes of hsa-miR-20a-3p,and perform GO functions on the target genes Annotation and enrichment analysis of KEGG signaling pathway through the DAVID database.Results:The mature base sequence of hsa-miR-20a-3p is highly conserved among various species;156 target gene sets are predicted;GO function annotation analysis results show that hsa-miR-20a-3p target genes are mainly enriched in 9 cell components such as nucleoplasm,cytoplasm and nucleus,are enriched in 11 molecular functions such as protein binding,chromatin binding,and DNA binding,and are enriched in 15 organisms such as forebrain morphogenesis,learning or memory,organ induction,gene expression regulation,etc.(P<0.05);KEGG signal pathway is significantly enriched in 3 signal pathways of virus carcinogenesis,hepatitis B and HTLV-Ⅰinfection(P<0.05).Conclusion:hsa-miR-20a-3p extensively participates in many important biological processes and signal pathways by regulating target genes,especially the process of viral carcinogenesis is a direction worthy of further study.
