莱菔硫烷联合维生素D对乳腺癌细胞增殖及凋亡的影响 被引量:2

摘要 目的:探究莱菔硫烷(SFN)联合维生素D(VD)对人乳腺癌MCF-7细胞增殖及凋亡的影响,为预防和控制乳腺癌提供新的策略。方法:实验分为空白对照组、SFN组、VD组和SFN+VD组。人乳腺癌MCF-7细胞培养至对数生长期,倒置显微镜、细胞血球板计数、CCK-8法和流式细胞术分别观察、分析和检测对照组和处理组干预MCF-7细胞后的细胞形态、数量、细胞增殖活力和细胞凋亡情况。结果:倒置显微镜下可以观察到,MCF-7细胞数量随药物浓度增加而逐渐减少,细胞形态也逐渐皱缩。细胞血球板计数结果显示,随着SFN或VD浓度的上升,人乳腺癌MCF-7细胞的数目随之下降。CCK-8检测结果表明,SFN和VD均可抑制MCF-7细胞增殖能力,且随药物浓度的增加,这种抑制效果更加显著;SFN+VD与两类药物单独作用相比,其对MCF-7细胞增殖的抑制效果更加显著,当药物浓度为10μmol/L时,SFN组、VD组和SFN+VD组中的MCF-7细胞增殖活力分别为对照组的47.54%、43.38%和32.29%,差异有统计学意义(均P<0.01)。流式细胞术检测结果显示,MCF-7细胞的凋亡比例随药物浓度的增加而显著上升,且SFN联合VD组最为显著。其中,对照组、10μmol/L的SFN组、VD组和SFN+VD处理组中的MCF-7细胞凋亡率分别为9.99%、33.51%、57.90%和90.83%,差异有统计学意义(均P<0.01)。结论:SFN联合VD能够加强对人乳腺癌MCF-7细胞生长的抑制作用,并诱导乳腺癌细胞凋亡。 Objective:To explore the effect of sulforaphane(SFN)combined with vitamin D(VD)on the proliferation and apoptosis of human MCF-7 breast cancer cells,and to provide new strategies for the prevention and control of breast cancer.Methods:The experiment consisted of blank control group,SFN treatment group,VD treatment group,and SFN+VD treatment group.MCF-7 cells were cultured to a logarithmic growth phase.The morphology,number,and cell proliferation activity and apoptosis of MCF-7 cells were observed,analyzed and detected by inverted microscope,counting plate,CCK-8 method and flow cytometry after intervention in control group and treatment groups.Results:Under inverted microscope,it can be observed that the number of MCF-7 cells decreased gradually with the increase of drug concentration,and the cell morphology also gradually shrank.The result of counting plate showed that the number of MCF-7 cells decreased with the increase of SFN or VD concentration.The result of CCK-8 test showed that SFN and VD could inhibit the proliferation of MCF-7 cells,and the inhibitory effect was more obvious with the increase of drug concentration.The inhibitory effect of SFN combined with VD on the proliferation of MCF-7 cells was more significant than that of the two substances alone.When the drug concentration was 10μmol/L,the proliferation activity of MCF-7 cells in SFN group,VD group and SFN+VD group were 47.54%,43.38%,and 32.29%of control group respectively,and the differences were statistically significant(all P<0.01).The result of flow cytometry showed that the percentage of apoptotic cells increased significantly with increasing drug concentration,especially in SFN+VD group.The apoptosis rates of MCF-7 cells in control group,10μmol/L SFN group,VD group and SFN+VD group were 9.99%,33.51%,57.90%and 90.83%respectively,and the differences were statistically significant(all P<0.01).Conclusion:SFN combined with VD can enhance the inhibitory effect on the growth of MCF-7 cells and induce apoptosis of MCF-7 cells.
