双氧化酶成熟因子2基因检测在肺癌患者淋巴结转移及预后预测中的价值研究

Value of DUOXA2 level in predicting lymph node metastasis and prognosis of lung cancer patients

目的:研究双氧化酶成熟因子2(DUOXA2)基因在肺癌患者肿瘤组织中的表达,以及其与肺癌患者临床病理特征和预后的相关性。方法:选取90例接受手术治疗的肺癌患者,通过定量PCR检测肺癌肿瘤组织和癌旁正常组织中DUOXA2 mRNA表达,通过免疫印迹法和免疫组织化学法检测DUOXA2蛋白表达,通过多因素回归分析法分析肺癌患者预后相关因素。结果:DUOXA2 mRNA和蛋白在90例肺癌患者肿瘤组织中的表达水平显著高于癌旁正常组织(P<0.05)。DUOXA2蛋白在肺癌肿瘤组织中的表达水平与肺癌患者年龄、性别、病理类型、肿瘤大小和TNM分期无关(均P>0.05),而与淋巴结是否转移有关(P<0.05);DUOXA2 mRNA和蛋白在30例淋巴结转移肺癌患者肿瘤组织中的表达水平显著高于其在60例淋巴结未转移肺癌患者肿瘤组织中的表达水平。多因素回归分析结果显示:DUOXA2在肺癌患者肿瘤组织中的表达水平是影响肺癌患者预后的独立危险因素(P=0.039)。结论:DUOXA2基因在肺癌患者肿瘤组织中表达升高,其表达水平与肺癌患者是否出现淋巴结转移有关,是影响肺癌患者预后的独立危险因素。

Objective:To study the expression of DUOXA2 gene in tumor tissues of patients with lung cancer and its correlation with clinicopathological characteristics and prognosis of patients with lung cancer.Methods:A total of 90 lung cancer patients who received surgical treatment were selected.The DUOXA2 mRNA expression in lung cancer tumor tissue and adjacent normal tissue was detected by quantitative PCR.DUOXA2 protein expression was detected by Western blot and immunohistochemical methods.The prognostic factors of lung cancer patients were analyzed by multivariate regression analysis.Results:The expression levels of DUOXA2 mRNA and protein in tumor tissues of 90 lung cancer patients were significantly higher than those in adjacent normal tissues(all P<0.05).The expression level of DUOXA2 protein in lung cancer tumor tissue was not related to age,gender,pathological type,tumor size and TNM stage of lung cancer patients(all P>0.05),but was related to lymph node metastasis(P<0.05).The expression levels of DUOXA2 mRNA and protein in 30 lung cancer patients with lymph node metastasis were significantly higher than those in 60 lung cancer patients without lymph node metastasis(P<0.05).Multivariate regression analysis showed that the expression level of DUOXA2 in tumor tissues of lung cancer patients was an independent risk factor for prognosis of lung cancer patients(P=0.039).Conclusion:The expression of DUOXA2 gene is increased in tumor tissues of lung cancer patients.The expression level of DUOXA2 gene is related to lymph node metastasis in lung cancer patients and is an independent risk factor affecting the prognosis of lung cancer patients.
