miR-125b、miR-126在儿童哮喘中的诊断价值及其与哮喘严重程度的关系 被引量:7

摘要 目的探讨微小核糖核酸-125b(miR-125b)、miR-126对儿童哮喘的诊断价值,分析二者与哮喘严重程度的关系。方法选取该院2017年9月至2020年12月收治的120例哮喘患儿、79例单纯上呼吸道感染患儿作为哮喘组、感染组,另选取同期84例体检健康志愿儿童作为对照组。检测并比较3组血清miR-125b、miR-126水平及肺功能指标[用力肺活量(FVC)、第1秒用力呼气容积(FEV_(1))、FEV_(1)/FVC],采用Pearson线性相关分析血清miR-125b、miR-126水平与肺功能指标的相关性,绘制受试者工作特征(ROC)曲线,分析miR-125b、miR-126单独和联合检测诊断哮喘的曲线下面积(AUC)。根据哮喘严重程度,将哮喘患儿分成轻度组、中度组及重度组,比较3组血清miR-125b、miR-126水平,采用Spearman相关分析血清miR-125b、miR-126水平与患儿哮喘严重程度的相关性。结果哮喘组血清miR-125b、miR-126水平高于感染组与对照组(P<0.05),感染组血清miR-125b、miR-126水平高于对照组(P<0.05);哮喘组FEV_(1)、FEV_(1)/FVC低于感染组与对照组(P<0.05)。血清miR-125b、miR-126水平与FEV_(1)、FEV_(1)/FVC均呈负相关(r<0,P<0.05)。血清miR-125b、miR-126单独和联合检测诊断哮喘的AUC分别为0.763、0.749、0.846。重度组、中度组血清miR-125b、miR-126水平高于轻度组(P<0.05),重度组高于中度组(P<0.05)。血清miR-125b、miR-126水平与哮喘严重程度均呈正相关(r>0,P<0.05)。结论哮喘患儿血清miR-125b、miR-126水平明显升高,二者对哮喘诊断及哮喘严重程度评估均有一定价值。 Objective To investigate the diagnostic value of microribonucleic acid-125b(miR-125b)and miR-126 in children with asthma,and analyze the relationship between the two factors and the severity of asthma.Methods A total of 120 children with asthma and 79 children with simple upper respiratory tract infection who were admitted in this hospital from September 2017 to December 2020 were selected as the asthma group and the infection group,and 84 volunteer children with healthy physical examination during the same period were selected as the control group.Serum levels of miR-125b,miR-126 and pulmonary function indicators[forced vital capacity(FVC),forced expiratory volume in 1 second(FEV_(1)),FEV_(1)/FVC]were detected and compared among the three groups.Pearson linear correlation was used to analyze the correlation between serum levels of miR-125b and miR-126 and lung function indicators.The receiver operating characteristic(ROC)curve was drawn to analyze the area under the curve(AUC)of miR-125b and miR-126 alone and in combination for the diagnosis of asthma.According to the severity of asthma,children with asthma were divided into the mild group,the moderate group and the severe group,and serum levels of miR-125b and miR-126 were compared among the three groups,and the correlation between the serum levels of miR-125b and miR-126 and the severity of asthma in children was analyzed by Spearman correlation.Results The serum levels of miR-125b and miR-126 in the asthma group were higher than those in the infection group and the control group(P<0.05),and the serum levels of miR-125b and miR-126 in the infection group were higher than those in the control group(P<0.05).The FEV_(1) and FEV_(1)/FVC in the asthma group were lower than those in the infection group and the control group(P<0.05).The serum levels of miR-125b and miR-126 were negatively correlated with FEV_(1) and FEV_(1)/FVC(r<0,P<0.05).The AUCs of serum miR-125b and miR-126 alone and in combination to diagnose asthma were 0.763,0.749 and 0.846,respectively.The serum levels of miR-125b and miR-126 in the severe group and the moderate group were higher than those in the mild group(P<0.05),and the serum levels in the severe group were higher than those in the moderate group(P<0.05).The serum levels of miR-125b and miR-126 were positively correlated with the severity of asthma(r>0,P<0.05).Conclusion The serum levels of miR-125b and miR-126 in children with asthma significantly increase.They all have a certain value in the diagnosis of asthma and assessment of asthma severity.
