姜黄素对重症手足口病模型大鼠的作用研究

摘要 目的:研究姜黄素对重症手足口病模型大鼠的作用。方法:从60只SD大鼠中随机选出50只通过腹腔注射EV71BJ毒株建立重症手足口病大鼠模型,余下10只作为对照组,将建模成功的大鼠随机分为模型组、阿昔洛韦组(6.6 mg·kg^(-1))及姜黄素高(20 mg·kg^(-1))、中(10 mg·kg^(-1))、低(5 mg·kg^(-1))剂量组,并灌胃给予相应药物,模型组和对照组灌胃等体积生理盐水,连续治疗2周。在治疗第1天、第4天、第7天、第10天、第13天记录大鼠体质量的变化;RT-qPCR检测大鼠肌肉组织EV71 BJ病毒载量;ELISA法检测大鼠血浆D-二聚体(D-dimer,D-D)、神经元特异性烯醇化酶(neuron-specific enolase,NSE)和神经肽Y(neuropeptide Y,NPY)的水平。结果:与第1天比较,第7天、第10天及第13天各给药组大鼠体质量显著升高(P<0.05);与第4天比较,第7天、第10天及第13天各给药组大鼠体质量显著升高(P<0.05),且呈时间依赖性;与对照组比较,模型组体质量显著下降(P<0.05),D-D、NSE、NPY的水平及EV71 BJ mRNA的水平显著升高(P<0.05);与模型组比较,第7天、第10天及第13天各给药组大鼠体质量均显著升高(P<0.05),各给药组D-D、NSE、NPY的水平及EV71 BJ mRNA的水平显著下降(P<0.05),且各给药组大鼠血浆D-D、NSE和NPY水平均呈时间依赖性降低(P<0.05)。结论:姜黄素可明显升高重症手足口病大鼠的体质量,降低病毒载量及血浆D-D、NSE、NPY水平。 Objective:To study the effect of curcumin on severe hand,foot and mouth disease model rats.Methods:Fifty SD rats were randomly selected from 60 SD rats to establish a rat model of severe hand,foot and mouth disease by intraperitoneal injection of EV71 BJ strain,and the remaining 10 were used as a control group.Acyclovir group(6.6 mg·kg^(-1))and curcumin high(20 mg·kg^(-1)),medium(10 mg·kg^(-1))and low(5 mg·kg^(-1))dose groups were given corresponding drugs by gavage,equal volume of normal saline was administered by gavage to model group and control group A,and the treatment was continued for 2 weeks.On the 1st,4th,7th and 13th days of treatment,the changes of body weight of rats were recorded;the EV71 BJ viral load in rat muscle tissue was detected by RT-qPCR;the levels of D-D,NSE and NPY in rat plasma were detected by ELISA.Results:Compared with the first day,the body weight of the rats in each administration group was significantly increased on the 7th,10th and 13th days(P<0.05);On the 13th day,the body weight of the rats in each administration group increased significantly(P<0.05);compared with the 7th day,the body weight of the rats in each administration group increased significantly on the 10th and 13th day(P<0.05);Compared with the 10th day,the body weight of the rats in each administration group was significantly increased on the 13th day(P<0.05);compared with the control group,the body weight of the model group was significantly decreased(P<0.05).D-D,NSE,NPY and EV71 The level of BJ mRNA was significantly increased(P<0.05);compared with the model group,the body weights of the rats in each administration group were significantly increased on the 7th,10th and 13th days(P<0.05).The levels of D-D,NSE,NPY,EV71 BJ mRNA in the groups decreased significantly(P<0.05),and the plasma D-D,NSE and NPY levels of the rats in each administration group decreased in a time-dependent manner(P<0.05).Conclusion:Curcumin can significantly reduce the weight loss of severe hand-foot-mouth disease rats,reduce the viral load and plasma levels of D-D,NSE and NPY to treat hand-foot-mouth disease.
