摘要
目的探讨纤维蛋白原(fibrinogen,Fib)及血小板膜糖蛋白(glycoprotein,GP)Ⅱb/Ⅲa基因多态性与家族聚集性大动脉粥样硬化性(large‑artery atherosclerosis,LAA)脑梗死的关系。方法收集家族聚集性LAA脑梗死和散发性LAA脑梗死患者各25例,分别为家族组及散发组,选择25例非心脑血管疾病患者作为对照组。75例患者均提取基因组DNA,以基因组DNA为模板对待检序列进行PCR反应,纯化PCR产物,对PCR产物直接进行测序,确定有无纤维蛋白原及血小板GP IIb/IIIa基因多态性存在。结果Fib基因多态性在3组间差异无统计学意义(P>0.05),GP IIb位点在3组之间差异有统计学意义(P<0.05),GPⅢa位点在家族组、散发组、对照组3组之间差异无统计学意义(P>0.05)。结论在家族聚集性LAA脑梗死中有GPⅡb基因多态性异常,而Fib及GPⅢa基因型无异常。
Objective:To investigate the relationship between fibrinogen(Fib)and glycoprotein(GP)Ⅱ b/Ⅲ a gene polymorphisms and familial aggregation large-artery atherosclerosis(LAA)cerebral infarction. Methods:Twenty-five patients with familial aggregation LAA cerebral infarction and 25 patients with sporadic LAA cerebral infarction were collected as family group and sporadic group respectively. Twenty-five healthy people were selected as the control group. Genomic DNA(g DNA)was extracted from the 75 cases,and the sequence to be tested was subjected to PCR reaction with g DNA as template. The PCR products were purified and sequenced directly to determine whether there were fibrinogen and platelet membrane glycoprotein Ⅱb IIIa gene polymorphisms. Results:There was no significant difference in Fib gene polymorphism among the three groups(P > 0. 05),there was significant difference in GP IIb locus among the three groups(P < 0. 05),and there was no significant difference in Gp Ⅲa locus among family group,sporadic group and control group. Conclusion:GP Ⅱb gene polymorphism is abnormal in familial aggregation LAA cerebral infarction,but Fib and Gp IIIa genotypes are not abnormal.
